摘 要:目的探讨腺病毒KLF7对大鼠脊髓半横断损伤后酪氨酸激酶A(Trk A)和酪氨酸激酶B(TrkB)表达、损伤灶形态及运动功能恢复的影响。方法制作大鼠脊髓半横断损伤模型,分别将腺病毒2(AAV2)、腺病毒2-KLF7(AAV2-KLF7)用微量注射器注射到脊髓损伤灶,实验大鼠随机分为正常组、AAV2对照组及AAV2-KLF7组。术后4周,通过HE染色观察脊髓损伤灶面积,Western Blot检测KLF7、TrkA和TrkB蛋白表达,BBB行为学评价运动功能恢复。结果与对照组比较,AAV2-KLF7组脊髓半离断损伤灶面积比显著缩小(P<0.05),KLF7、Trk A、Trk B蛋白相对表达显著增高(P<0.05)。AAV2-KLF7组BBB评分显著高于对照组(P<0.05)。结论 KLF7通过上调神经营养因子受体Trk A和Trk B表达,改善大鼠脊髓损伤灶形态,促进运动功能的恢复。Objective To study the effect of adenovirus KLF7 on the expression of tyrosine kinase A (Trk A) and tyrosine ki- nase B (Trk B), and lesion morphology and motor function recovery in the rat with spinal cord injury. Methods The rat spinal cord semi transected model was made. Adenovirus2 ( AAV2), adenovirus2 KLF7 ( AAV2 - KLF7 ) with a micro syringe injection into the spinal cord lesion, the rats were randomly divided into normal group, AAV2 control group and AAV2 - KLF7 group. Four weeks later, the lesion area of spinal cord injury was observed by HE staining. KLF7, Trk A and TrkB protein expression was detected by Western Blot, and the neurological functions were evaluated using the Basso - Beattie - Bresnahan (BBB) scale. Results Compared with the control group, the lesion area in AAV2 - KLF7 group was significantly reduced (P 〈 0.05 ), and the protein expression of KLFT, Trk A and Trk B were significantly increased (P 〈 0.05). The BBB score in AAV2 - KLF7 group was significantly higher than that in con- trol group (P 〈 0.05). Conclusion KLF7 could improve the rat spinal cord injury lesion as well as motor function recovery, which may be relating with the upregulating expression of neurotrophic factor receptor Trk A and Trk B.
