摘 要:目的:观察参松养心胶囊对血管紧张素Ⅱ(AngⅡ)引起的心肌细胞凋亡的影响。方法:分离大鼠乳鼠心肌细胞,随机分为对照组(PBS)、AngⅡ(0.1μmol·L^(-1))组、参松养心胶囊(10μmol·L^(-1))预处理1 h+AngⅡ组;Mn(Ⅲ)TBAP(2.5μmol·L^(-1))预处理1 h+AngⅡ组。通过MTT和Caspase-3/7活性检测试剂盒检测细胞活性和细胞凋亡变化;利用western-blot检测心肌细胞SOD-1和Nox2/gp91phox表达。结果:与对照组比较,AngⅡ引起心肌细胞活性明显下降,凋亡细胞显著增加,心肌细胞SOD-1表达下调,而Nox2/gp91phox表达上调。参松养心胶囊预处理后,再给予AngⅡ干预,与AngⅡ处理组比较,心肌细胞活性明显增加,凋亡细胞显著减少,心肌细胞SOD-1表达上调,而Nox2/gp91phox表达下调。Mn(Ⅲ)TBAP作为超氧自由基的清除剂,与参松养心胶囊胶囊作用类似,同样可以抑制AngⅡ引起的细胞损伤。结论:参松养心胶囊通过抑制心肌细胞氧化应激反应,减轻AngⅡ引起的细胞损伤。Objective: To observe the influence of Shensong Yangxin Capsules( SSYXC) on myocardial cell apoptosis due to angiotensin Ⅱ. Methods: The chosen myocardial cells of infantile rats were separated,and were randomly divided into the control group( PBS),the Ang Ⅱ( 0. 1 μmol·L^-1) group,SSYXC( 10 μmol·L^-1) pretreatment + Ang Ⅱgroup,the Mn(Ⅲ) TBAP( 2. 5μmol·L^-1) pretreatment 1 h + Ang Ⅱ group. MTT and Caspase-3/7 activity detection kit,the changes of cell viability and apoptosis were detected. By using western-blot,the expression of SOD-1 and Nox2/gp91 phox were detected. Results: Compared with the control group,Ang Ⅱ significantly decreased the activity of myocardial cells,the apoptotic cells increased significantly,the expression of SOD-1 was down-regulated,and the expression of Nox2/gp91 phoxwas up-regulated. After pretreatment with SSYXC plus Ang Ⅱ intervention,compared with the Ang II treatment group,the activity of myocardial cells increased,apoptotic cells decreased significantly; the expression of SOD-1 was up-regulated,while that of Nox2/gp91 phoxwas down-regulated. Mn( Ⅲ)TBAP as superoxide radical scavenger,it has similar function to that of SSYXC,can also inhibit the cell injury caused by Ang Ⅱ.Conclusion: SSYXC can reduce the cell damage caused by Ang II by inhibiting the oxidative stress in myocardial cells.
