摘  要:目的:探讨加味四物汤水提物(mSWT)对急性心肌梗死(AMI)模型小鼠骨髓细胞动员及定植的影响。方法:结扎昆明种小鼠冠状动脉左前降支,制备AMI模型;DAPI胫骨骨髓腔注射标记骨髓细胞,灌胃给予mSWT,流式细胞术检测外周血EPCs比例;心肌组织病理学观察骨髓细胞在AMI区的定植和微小血管密度。结果:与模型组比较,60 mg/kg mSWT治疗后,mSWT治疗组外周血中EPCs细胞比例明显增加(P<0.05);AMI小鼠心肌梗死面积明显减小,梗死区炎性细胞浸润较少,心肌细胞变性坏死减轻,存活的心肌组织较多,明显可见较多的DAPI标记骨髓来源细胞在AMI区域定植、存活,且新生毛细血管增多。结论:mSWT能明显提高AMI小鼠外周血EPCs细胞比例,改善AMI小鼠心肌组织病理改变,其作用可能与动员骨髓细胞趋化至AMI区域定植,促进血管新生有关。Objective To investigate the effects of Modified 5iwutang aqueous extract (mSWT) on bone marrow cell mobilization and plantation in mice after acute myocardial infarction (AMI). Methods The AMI model was established by ligating the left anterior descending artery in Kunming mice. Bone marrow cells were marked by injection of DAPI into the cavitas medullaris of shin bones. Mice were administered with mSWT 60 mg/kg by intragastric administration once a day after surgery. After haematoxylin-eosin and immunofluorescent staining, the plantation of bone marrow cells in AMI area and mierovessel density were detected by histopathological assay. The EPCs proportion of peripheral blood was measured by flow cytometry. Results Compared with model mice, infarct size obviously was decreased after mSWT 60 mg/kg treatment, with EPCs ratio increased significantly. Inflammatopy cell infiltration of myocardium was much less. Myocardium cellular degeneration and necrosis , fibrosis were reduced, which survival cardiac muscle tissue was increased. DAPI positive cells of plantation and survive were found in AMI region of mSWT group. The number of positive cells were increased in mSWT group than that of model group. There were lot of microvessels of vWF positive expresseion in mSWT than model group. Conclusion : mSWT significantly improved histopathology of AMI mice. The mechanism may be involved in moved bone marrow cells , homed to AMI area and promote angiogenesis.
