摘 要:目的探讨补充维生素D_3(vitamin D_3,VD_3)的同时补充维生素C(vitamin C,VC)是否可以促进维生素D_3活性形式1,25-(OH)_2D_3的生成,从而改善豚鼠溃疡性结肠炎。方法雄性Dunkin-Hartley豚鼠56只,按体重随机分为对照组、低剂量VD_3低剂量VC组、低剂量VD_3中剂量VC组、低剂量VD_3高剂量VC组、高剂量VD_3低剂量VC组、高剂量VD_3中剂量VC组、高剂量VD_3高剂量VC组共7组。维生素D_3和维生素C干预7周后,除对照组外,其余6组自由饮用3%葡聚糖硫酸钠溶液3 d,建立溃疡性结肠炎的模型。造模期间进行疾病活动指数评分,造模结束后取豚鼠血清和结肠组织进行指标检测:ELISA方法检测豚鼠血清中25(OH)D_3、1,25-(OH)_2D_3、钙离子、血清碱性磷酸酶、肿瘤坏死因子α水平;生化法检测结肠组织中髓过氧化物酶活性。结果当维生素D_3水平一定时,豚鼠疾病活动指数评分、髓过氧化物酶活性和肿瘤坏死因子α水平随维生素C的增高明显降低,25(OH)D_3、1,25-(OH)_2D_3水平明显增高,血清碱性磷酸酶和血清钙增高。结论维生素C和维生素D_3联合应用可以改善豚鼠溃疡性结肠炎,可能的机制是维生素C促进维生素D_3活性形式的生成。Objective To explore whether the supplementation of vitamin 93 and vitamin C can promote the formation of 1,25- (OH)2D3, the active form of vitamin D-, and reducing the development of ulcerative colitis. Methods Fifty-six adult male Dunkin-Hartley guinea pigs were randomly divided into seven groups: the control group, low dose of vitamin D3 and low dose of vitamin C, low dose of vitamin D3 and moderate dose of vitamin C, low dose of vitamin D3 and high dose of vitamin C, high dose of vitamin D3 and low dose of vitamin C, high dose of vitamin D3 and moderate dose of vitamin C, high dose of vitamin D3 and high dose of vitamin C. After seven weeks of vitamin D3 and vitamin C intervention, the guinea pigs were gavaged with 3% dextran sulfate solution for three clays to establish experimental models for ulcerative colitis, except for the control group. After modeling, the guinea pigs were sacrificed, and their serum and colon tissues were collected. The disease activity index was scored during modeling. The level of 25 (OH)D3, 1,25-(OH)2D3, calcium ion, alkaline phosphatase and TNF-α in the serum were measured by ELISA. The MPO activity of colon tissue was detected by biochemical method. Results The scores of DAI, the activity of MPO and TNF-α decreased significantly with the content of vitamin C increased when the vitamin D3 level was constant. The level of 25 (OH)D3,1,25-(OH)2D3, alkaline phosphatase and serum calcium was significantly increased. Conclusion Vitamin C in combination with vitamin D3 can meliorate the inflammation of ulcerative colitis, and the possible mechanism may be that vitamin C can promote the formation of 1,25-(OH)2D3.
