摘  要:目的探讨铁过载对雄性小鼠心肌组织JWA基因m RNA表达的影响及其与氧化应激损伤的相关性。方法 40只雄性小鼠按体重随机分成4组(对照组和3个铁过载组),各组隔天腹腔注射不同剂量(0、50、100、150 mg/kg·wb)的右旋糖苷铁,持续注射4周后,眼球取血测定血清铁;处死小鼠,取肝脏组织测肝组织铁,取心肌组织,RT-PCR技术测心肌组织JWA基因mRNA表达量,并测心肌组织丙二醛(MDA)、过氧化氢酶(CAT)、过氧化氢(H_2O_2)、羟自由基和谷胱甘肽过氧化物酶(GSH-PX)含量。结果与对照组相比,3个过载组血清铁及肝组织铁含量均显著升高(P值均<0.05);100 mg/kg·wb剂量组、150 mg/kg·wb剂量组铁过载小鼠心肌组织的JWA基因mRNA表达水平显著升高,差异均有统计学意义(P<0.05)。心肌组织JWA基因mRNA表达量与其CAT活力、过氧化氢含量、GSH-PX水平和羟自由基活力呈负相关关系,而与MDA含量呈正相关关系(P<0.05)。氧化应激水平显示,与对照组相比,3个铁过载组心肌匀浆MDA含量均升高(P<0.05);而CAT活力、H_2O_2含量、羟自由基活力、GSH-PX均降低(P<0.05)。结论铁过载导致小鼠心肌组织JWA基因mRNA高表达,并与心肌组织氧化应激损伤密切相关。Objective The aim of this study was to investigate the effect of iron overload on the expression of JWA mRNA in myocardium of male mice and its relationship with oxidative stress injury. Methods Forty male mice were randomly divided into four groups (control group and three iron overload groups) according to their body weight. Each group was injected intraperitoneally with different doses (0mg/kg.bw, 50mg/kg.bw, 100mg/kg.bw, 150mg/kg.bw). After four weeks of continuous injection, serum iron was measured in the eyeballs. The mice were sacrificed and liver tissue was taken for determination of hepatic iron. The expression of JWA mRNA was measured by RT-PCR. MDA, catalase (CAT), hydrogen peroxide (H202), hydroxyl free radical and glutathione peroxidase (GSH-PX) in myocardium were measured. Results Compared with the control group, iron content in serum and hepatic tissue of three overload groups were significantly increased (P〈0.05). The myocardial tissue of iron overload mice in the dose of 100mg/kg.bw and 150mg/kg.bw (P〈0.05). The expression of JWA gene was significantly higher than that of the control group (P〈0.05). There was a negative correlation between the expression of JWA mRNA and the activity of CAT, H202, GSH-PX and hydroxyl free radical in myocardium, but positively correlated with MDA content (P〈0.05). Compared with the control group, the levels of MDA, MDA, CAT activity, H2O2, hydroxyl free radical and GSH-PX in the three iron overload groups were significantly decreased (P〈0.05). Conclusion Iron overload resulted in high expression of JWA mRNA in myocardium of mice, and was closely related to myocardial oxidative stress injury.
