IL-10、TNF-α及SOCS3在妊娠期肝内胆汁淤积症母胎界面的变化

摘　　要：目的探讨白细胞介素-10(IL-10)、肿瘤坏死因子-α(TNF-α)及细胞因子信号转导抑制子3(SOCS3)在妊娠期肝内胆汁淤积症(ICP)母胎界面的变化。方法选取新疆医科大学第五附属医院2013年10月至2016年10月ICP孕妇43例作为观察组,选择同期产检的健康孕妇47例作为对照组。采用Envision免疫组化法测定两组孕妇胎盘组织中IL-10、TNF-α的表达水平,采用酶联免疫吸附测定法(ELISA)和蛋白免疫印迹法(Western-blot)测定两组孕妇胎盘组织中SOCS3的表达水平。结果观察组IL-10的表达明显低于对照组,差异具有统计学意义(Z=-2.578,P<0.05),TNF-α的表达明显高于对照组,差异有统计学意义(Z=-4.421,P<0.05);观察组的孕妇胎盘组织中SOCS3表达水平明显低于对照组,差异有统计学意义(Z=-2.325,P<0.05);两组患者SOCS3的OD值比较差异有统计学意义(t=565.702,P<0.05)。结论 ICP患者的母胎界面TNF-α表达增加,IL-10表达降低,胎盘组织中SOCS3参与了ICP的发病。Objective To study the changes of interleukin 10（ IL-10）,tumor necrosis factor α（ TNF-α） and suppressor of cytokine signaling 3（ SOCS3） at maternal-fetal interface of patients with intrahepatic cholestasis of pregnancy（ ICP）. Methods From October 2013 to October 2016,43 pregnant women with ICP in the Fifth Affiliated Hospital of Xinjiang Medical University were selected in observation group,and 47 healthy pregnant women taking prenatal examination in the same period were selected in control group. The expressions of IL-10 and TNF-α level in placenta of two groups were determined by Envision immunohistochemical method,and the expression level of SOCS3 was determined by ELISA method and Western blot（ Western-blot） method. Results The expression of IL-10 of the observation group was significantly lower than the control group with statistical significance（ Z =-2. 578,P〈 0. 05）,but the expression of TNF-αwas significantly higher（ Z =-4. 421,P 〈0. 05）. The expression of SOCS3 of the observation group was significantly lower than the control group with statistical significance（ Z =-2. 325,P 〈0. 05）. There was obvious difference in OD value of SOCS3 between two groups（ t = 565. 702,P 〈0. 05）. Conclusion The expression of TNF-α increases but IL-10 decreases in maternal-fetal interface of patients with ICP,and SOCS3 in placenta is involved in the pathogenesis of ICP.
