摘  要:目的观察不同剂量LBP对大鼠视神经损伤后视神经修复的能力及对视网膜中SOD活性和MDA含量的影响。方法取SPF级健康雄性SD大鼠24只,随机分为正常组、损伤组、LBP低剂量组[100mg·(kg·d)^(-1)]和LBP高剂量组[400mg·(kg·d)^(-1)],每组各6只。除正常组外,其余3组均用微型视神经夹造成大鼠左眼视神经损伤模型。造模成功后正常组和损伤组大鼠每日生理盐水定时定量灌胃,LBP低剂量组和LBP高剂量组每日枸杞多糖定时定量灌胃,连续3个月。3个月后电镜下观察视神经超微结构的变化,并检测视网膜中SOD的活性和MDA的含量。结果损伤组视神经超微结构紊乱,轴索消失,髓鞘退变。与正常对照组相比,LBP低剂量组、高剂量组大鼠视神经轴索及髓鞘等出现一定的形态学改变,但明显轻于损伤组。高剂量组视神经形态学类似于正常对照组,明显优于低剂量组。视网膜中SOD、MDA含量四组间比较差异均有统计学意义(P<0.05)。损伤组、枸杞多糖低剂量组和枸杞多糖高剂量组SOD含量均低于正常组,MDA均高于正常组(P均<0.05);枸杞多糖低剂量组和高剂量组中SOD的含量均高于损伤组,MDA均低于损伤组(P均<0.05);枸杞多糖高剂量组SOD的含量高于低剂量组,MDA含量低于低剂量组(P均<0.05)。结论 LBP可以修复视神经损伤造成的视神经超微结构的改变,提高视网膜中SOD活性,减少MDA生成,从而保护视网膜神经节细胞。Objective to observe self-repairing ability of different doses of Lycium barbarucham polysacride (LBP) on optic nerve and effect of LBP on super oxidedismutase (SOD) and malondiadehyde (MDA) in the injured optic nerve rats model. Methods 24 SPF male SD rats were divided into four groups randomly, the normal group, the injured group, the low doses of LBP 100mg·(kg·d)^-1 group and the high doses of LBP 400mg·(kg·d)^-1 group. Except the normal group, the others used mini-clamp to make injured optic nerve rats model on the left eye. the rats in the normal group and the injured group were fed on equal saline at regular time every day. The rats in the low doses of LBP group and the high doses of LBP group were fed on LBP 100mg·(kg·d)^-1 and 400mg ·(kg·d)^-1 respectively every day. The rats were sacrificed at the 3rd month and the contents of SOD and MDA in retina were detected. Results Electron microscope showed the optic nerve ultrastructural disorder, axon disappearance, myelin degeneration in the injured group. Compared with the normal control group, axon and myelin of the optic nerve in LBP low dose group and high dose group showed some morphological changes and it was significantly lighter than the injury group. The optic nerve morphology of the high dose group was similar to that of the normal control group, which was obviously superior to the low dose group. The SOD and MDA contents in the retina were significantly different among the four groups. The contents of SOD in injured group, LBP low dose group and high dose group were significantly lower than that in normal group (P〈0.05) while MDA contents in injured group, LBP low dose group and high dose group were significantly higher than that of the normal group (P〈0.05). The content of SOD in LBP low dose group and high dose group were significantly higher than that in injured group (P〈0.05) while MDA were lower than that in the injured group.
