摘  要:目的探讨神经营养因子BDNF、神经肽VGF及MAPK/Erk信号传导通路表达情况与海湾战争综合征(GWS)发病机制的关系。方法将36只SD大鼠随机分为对照组、限制应激组及GWS模型组,采用Abdel-Rahman的方法建立GWS大鼠模型。分别在0、7、14、21、28 d测定大鼠体质量。实验28 d结束后,处死实验大鼠、取脑、分离海马。采用免疫组化染色、VGF/MAPK免疫荧光双标染色及蛋白质印迹法(Western blot)检测VGF、BDNF、MAPK的表达情况。结果单因素方差分析3组大鼠体质量,组间比较差异有统计学意义(P<0.01)。GWS模型组大鼠体质量增长率明显低于对照组与限制应激组,差异有统计学意义(P<0.05)。海马CA1区VGF、BDNF、MAPK免疫组化染色的平均光密度值,限制应激组、GWS模型组较对照组均降低,差异有统计学意义(P<0.05);GWS模型组较限制应激组均降低,差异有统计学意义(P<0.05)。免疫荧光检测结果显示,海马CA3区细胞内MAPK与VGF的表达部位相同、变化趋势一致,MAPK、VGF荧光的表达与CA1区免疫组化结果一致。Western blot结果显示,限制应激组、GWS模型组与对照组比较,海马VGF、BDNF和MAPK的积分光密度值降低,差异有统计学意义(P<0.05);GWS模型组与限制应激组比较,海马VGF、BDNF和MAPK的积分光密度值降低,差异有统计学意义(P<0.05)。结论 GWS的发病机制与MAPK/ERK信号传导通路的抑制及VGF的表达减少相关。Objective To investigate neurotrophic factor of BDNF,neuropeptide VGF and MAPK/ERK signaling pathway abnormalities associated with pathogenesis of Gulf War syndrome( GWS). Methods A total of 36 SD rats were randomly divided into the control group,the restrained-stress,and GWS model group according to the Abdel-Rahman model establishing method. The body weight of rats were measured on 0,7,14,21 and 28 days. At the end of 28 days experiment,rats were killed,the brains were removed,and hippocampus was separated. The expressions of VGF,BDNF and MAPK in rat hippocampus were examined using SABC immunohistochemical staining,immunofluorescence double staining and western blot. Results Single-factor analysis of variance of 3 groups of rats body weight,there were statistically significant differences( P〈0. 01). The body weight growth rate in GWS model group was significantly lower than that in the control group and the restrained-stress group( P〈0. 05). Compared to the control group,the mean optical density of the VGF,BDNF and MAPK immunohistochemical staining in rat hippocampal CA1 region reduced in the GWS model group and the restrained-stress group( P〈0. 05);compared to the restrained-stress group,the mean optical density of the VGF,BDNF and MAPK immunohistochemical staining in rat hippocampal CA1 region reduced in the GWS model group( P〈0. 05). The MAPK expression showed the same location and trend as VGF expression in the hippocampal CA3 region using double immunofluorescence staining. Both MAPK and VGF expression in the hippocampal CA3 region were consistent with the immunohistochemical results of CA1 region. As Western blot analysis showed,the integral optical density values of VGF,BDNF and MAPK significantly reduced in both restrainedstress group and GWS model group compared with those in the control group( P〈0. 05);the integral optical density values of VGF,BDNF and MAPK significantly reduced in the GWS model group compared to the restrained-stress group( P〈0. 05).Conclusion
