结合静脉造影观察研究大鼠深静脉血栓的变化规律

摘　　要：目的建立SD大鼠深静脉血栓形成(DVT)模型,观察研究大鼠血栓模型在术前及术后不同时间进行下腔静脉造影,探讨血栓随时间变化的规律及意义。方法将23只SD大鼠切开皮肤及组织,暴露并游离下腔静脉,采用结扎阻断下腔静脉及其分支,并用微血管夹钳夹损伤静脉壁的方法诱导下腔静脉血栓形成;分别于术前第1天及术后第1、4、7、10、14、21天进行血管造影观察下腔静脉血栓变化及血管再通情况。结果最终17只大鼠参与实验全过程,术前第一天大鼠静脉造影显示下腔静脉通畅,未见充盈缺损。术后大鼠的血栓长度随时间的变化呈抛物线变化,血栓长度先增长,之后又开始减少,到术后第21天几乎溶解消失。结论经大鼠尾静脉行造影可成功观察到大鼠下腔静脉通畅情况及血管分支走行,是一种观察研究血栓变化的新的有效的实验方法。大鼠下腔静脉结扎后,很快就有生殖静脉代偿性增粗,参与静脉回流,不会引起严重的静脉栓塞症状,形成的下腔静脉血栓不经治疗一段时间后也可自溶。Objective To establish SD rat model of deep vein thrombosis, and to evaluate the significance of the dynamic changes of thrombus in rats before and after modeling operation with combination of inferior vena cava venography. Methods A total of 23 SD rats were taken in the test. The infrarenal portion of inferior vena cava of each rat was exposed through a midline laparotomy, venous thrombosis was induced in the inferior vena cava in a surgical procedure, which involved a combination of blocked blood flow after ligation and endothelial damage with the application of microvascular clip. Venography was performed respectively 1 day before operation and postoperative 1,4, 7, 10, 14, 21 days to observe the changes of thrombus and condition of vascular recanalization in rats. Results At last, 17 rats were survived during the whole experiment. The inferior vena cava was unobstructed, and no filling defects were found before the operation. Thrombus length changed with time in a curve of parabola in postoperative. The length of the thrombus inereasesd first and then decreasesd, almost completely dissolving after 21 days. Conclusion Venography through the rat caudal vein is a new method to observe the changes in inferior vena cava thrombus. Rats after ligation of the inferior vena cava have genital veins compensatory thickening in venous returnsoon, which will not cause serious symptoms of venous thrombosis, and the inferior vena cava thrombosis can also be autolysis without treatment after a period of time.
