摘 要:采用乙醇浸提法提取黑麦类黄酮物质,在单因素试验基础上,确定浸提溶剂乙醇体积分数为90%。进一步采用3因素2次正交回归设计,通过SAS软件RSREG过程分析乙醇用量、提取温度、提取时间对粗类黄酮得率的影响并优化工艺参数。结果表明,3因素对粗类黄酮得率的影响符合Y=B_0+∑B_jX_j+∑B_(ij)X_iX_j+∑B_(jj)X^2的3元2次回归模型,其关键参数主效应为乙醇用量(X_1)>提取温度(X_2)>提取时间(X_3);交互效应为X_1X_3>X_1X_2>X_2X_3;最优工艺参数为:乙醇用量71 mL(即料液比1∶14.2 g/mL)、提取温度58℃、提取时间176 min时,类黄酮得率94.23 mg/g。富含类黄酮的抽提物清除亚硝酸盐的效果很显著,清除率可达86.87%。Flavonoids were extracted from rye seed by using alcohol.Alcohol concentration of 90% was ascertained on the basis of four single-factor experiments.The optimal extraction conditions,including alcohol volume,extraction temperature and extraction time,were investigated on the basis of three factors quadratic orthogonal design to analyze the major effect and mutual effect between two factors on the yield of flavonoids by the RSREG process of SAS procedure.The results indicated that the factor effect on the yield of rye flavonoids was accord with the model of Y=B0+∑BjXj+∑B(ij)XiXj+∑B(jj)X^2,the major effect was the ratio of material and solvent(X1) temperature(X2) time(X3);the mutual effect between two factors was X2X3X1X2X1X3;the optimal technique parameters were as followed:alcohol volume of 71 ml,extraction temperature of 58 ℃ and extraction time of 176 min.Under above conditions,the extraction yield of rye flavonoids reached 94.23 mg/g.The extract showed powerful scavenging activities against nitrite.The scavenging rate reached 86.87%.
