九味肝泰对小鼠感染乙型肝炎病毒后肝硬化模型的干预研究  被引量:2

摘  要:目的观察九味肝泰对小鼠感染乙型肝炎病毒(hepatitis B,HBV)后肝硬化动物模型的影响及其干预机制。方法将48只BABL/c J小鼠随机均分为模型组和九味肝泰组,采用高压水注射方法将HBV质粒(50μL)通过尾静脉注入小鼠体内,然后腹腔注射50%CCI4(5 m L/kg)制备HBV肝硬化动物模型,另设12只为空白对照组;九味肝泰组用10%九味肝泰0.5 m L/(10 g.d)灌胃干预性治疗21 d,对照组和模型组灌等体积生理盐水。检测成模后和治疗21 d血清转化生长因子-βl(TGF-βl)、超氧化物歧化酶(SOD)、还原型谷胱甘肽(GSH)和肝组织中丙二醛(MDA)水平,并比较血清肝纤维化指标和肝功能。结果干预性治疗21 d后,九味肝泰组血清肝纤维化指标中血清IV型胶原(c IV)、Ⅲ型胶原(PC-III)、层粘连蛋白(LN)和透明质酸(HA)均明显降低,且TGF-βl和HBV DNA定量显著降低,肝功中谷草转氨酶(AST)、胆红素(TBIL)、谷氨酰转移酶(GGT)、谷丙转氨酶(ALT)和总胆汁酸(TBA)接近正常水平,肝组织中MDA明显降低,且静脉血GSH和SOD提高,与模型组和本组成模后比较,P<0.05。结论九味肝泰在抑制HBV DNA复制、减轻肝组织纤维化和促进肝功能恢复方面有明确的干预作用。Objective To observe the effects and intervention mechanism of nine kinds of liver and Thailand on the liver cirrhosis model of mice infected with hepatitis B virus (B hepatitis, HBV). Methods The 48 BABL / cJ mice were randomly divided into model group and nine flavour liver Thai group, using high pressure water injection methods HBV plasmid (50 μL) through the tail vein injection in mice, intraperitoneal injection of 50%CCI4 (5mL/kg) by HBV cirrhosis animal model, another 12 rabbits as blank control group; Jiuwei Gantai group with 10% Jiuwei liver Thai 0.5mL/(10g·d) irrigation stomach intervention treatment for 21 days (d), control group and model group were given equal volume of saline. Detection before and after the intervention of seroconversion growth factor beta 1 (TGF - β1, superoxide dismutase (SOD), also reduced glutathione (GSH) and liver malondialdehyde (MDA), compared with intervention before and after serum liver fibrosis markers and liver function. Results The intervention for the treatment of 21, Jiuwei Gantai group serum liver fibrosis indexes in serum type IV collagen (clV), type III collagen (pc-Ⅲ), layer adhesion protein (LN) and hyaluronic acid (HA) were significantly decreased, and TGF β1 and HBV DNA quantitative significantly decreased, liver function of aspertate transamination enzyme (AST), total bilirubin (TBIL), valley aminoacyl transferase (GGT), alanine aminotransferase (ALT) and total bile acid (TBA) close to the normal level, liver tissue MDA significantly reduced venous blood GSH and SOD increased, and the treatment group and model group,P〈0.05. Conclusion Nine kinds of liver and Thailand in inhibiting DNA HBV replication, reduce liver fibrosis and promote the recovery of liver function has a clear role in the intervention.
