摘  要:目的观察山茱萸多糖对亚急性衰老大鼠下丘脑-腺垂体-性腺轴的影响。方法 2014年3—6月开展实验,取雄性SD大鼠40只,随机数字表法分为对照组、模型组、山茱萸多糖组、雄激素组,每组10只。采用D-半乳糖(200mg·kg^(-1)·d^(-1))腹腔注射6周建立大鼠亚急性衰老模型,同时对照组、模型组每天给予生理盐水,药物组分别给予山茱萸多糖(50 mg·kg^(-1)·d^(-1))灌胃、雄激素(5 mg·kg^(-1)·d^(-1))肌肉注射6周。放射免疫(放免)法检测下丘脑促性腺激素释放激素(gonadotropin-releasing hormone,Gn RH)、卵泡刺激素(follicle stimulating hormone,FSH)、黄体生成素(luteotropic hormone,LH)、睾酮(testosterone,T)的含量。免疫组织化学法检测睾丸支持细胞雄激素结合蛋白(androgen binding protein,ABP)表达情况。计量资料多组间比较用方差分析,组间两两比较用LSD-t检验,P<0.05为差异有统计学意义。结果山茱萸多糖组Gn RH、FSH、LH水平较模型组明显下降,T水平明显升高,ABP表达的光密度水平明显增强,对比差异均有统计学意义(均P<0.05)。结论山茱萸多糖对衰老大鼠睾丸功能具有改善作用,其可能的机制与改善下丘脑-腺垂体-性腺轴的功能、增加睾丸组织ABP的表达有关。Objective To observe the effects of fructus corni polysaccharide on hypothalamus-pituitary-gonadal axis of the aging model rats.Methods The experiment was carried out from March to June 2014.40 male SD rats were randomly divided into four groups:the normal control group,model group,FCP group,androgen group.D-galactose was injected into the rats abdominal cavity by 200 mg·kg^-1·d^-1 for 6 weeks.At the same time,rats in FCP group were orally administered with FCP 50 mg·kg^-1·d^-1,rats in normal control group and model groups with normal saline,rats in androgen group intramuscular injection of androgen 5 mg·kg^-1·d^-1,for 6 weeks, gonadatropin releasing hormone(GnRH),follicle stimulating hormone(FSH),luteotropic hormone (LH) and testosterone(T) were detected by the method of radioimmunoassay.Observed the expression of androgen binding protein(ABP)in the testis.The difference between the groups was analyzed by one way ANOVA. The LSD-t test was compared between the groups, P〈0.05 was statistically significant. Results GnRH,FSH and LH of FCP group rats decreased significantly,T of FCP group rats increased significantly. The expression of ABP in FCP group rats was significantly enhanced (all P〈0.05). Conclusion FCP could improve the testicular function in aging rats,which may be related to prevent the dysfunction of hypothalamus-pituitary-gonadal axis,and enhance the expression of ABP in testicles.
