大鼠前体脂肪细胞分离培养方法的改进

摘　　要：前体脂肪细胞分离与培养是体外研究脂肪形成及其代谢机制的基础。本研究对目前采用的大鼠前体脂肪细胞分离培养方法进行了改进。前体脂肪细胞消化分离过程中,在Ⅰ型胶原酶消化后,取消尼龙筛过滤和离心操作,简化操作流程,减小污染几率,降低对前体脂肪细胞的损伤;加入研磨珠震摇,以充分分离前体脂肪细胞,提高细胞的获取率。改进后的前体脂肪细胞分离培养方法所培养的细胞生长曲线呈S型,符合细胞体外生长规律。本研究通过对操作过程的改良,建立了一种快速、方便、可靠的大鼠前体脂肪细胞分离培养体系。To establish a better method for isolating and culturing preadipocytes from rat white adipose tissue. The adiposetissue was digested by collagen, and after that the process of centrifugation and filtration were passed over. Therefore, the reducedprocessing of isolating preadipocytes decreased the probability of pollutions and cell trauma. After digestion, the ballotini wereadded to strike the digested tissue. Compared to the traditional method, the cell number obtained by the improved method wasincreased. The preadipocytes obtained by the improved method showed an S-shaped curve, which was accordance with the cellgrowth characteristics in vitro. The obtained cell could differentiate into adipocytes. It has been established an efficientpreadipocytes culture method which is easy, fast and reliable to isolate the preadipocytes.
