摘 要:目的 探讨草叶马尾藻多糖(polysaccharides from Sargassum graminifolium,SGP)影响肥胖小鼠肠道菌群的变化特征及其与血脂生化指标的关联性。方法 60只c57BL/6J小鼠,按数字表法随机分成普通饲料对照组(简称对照组,n=10)和高脂饲料组(n=50),高脂饲料组建立肥胖模型,剔除10只不符合标准的小鼠,剩余40只分为空白组和处理组,处理组分别以50 mg/kg(低剂量处理组)、100 mg/kg(中剂量处理组)、200 mg/kg(高剂量处理组)灌胃SGP,每组10只,持续8周,检测血脂生化指标三酰甘油(triglyceride, TG)、总胆固醇(total cholesterol, TC)、低密度脂蛋白(low density lopoprotein, LDL)和高密度脂蛋白(high density lopoprotein, HDL)含量和小鼠盲肠内容物的大肠杆菌、乳酸杆菌与双歧杆菌的菌落数。结果 空白组乳酸杆菌[(7.05±0.15)lg CFU/g]和双歧杆菌数量[(8.75±0.41)lg CFU/g]与对照组[乳酸杆菌(8.31±0.12)lg CFU/g、双歧杆菌(10.17±0.31)lg CFU/g]比显著减少,空白组大肠杆菌数量[(6.87±0.25)lg CFU/g]比对照组[(5.83±0.28)lg CFU/g]显著增加,差异均有统计学意义(P〈0.05)。与空白组相比,中、高剂量处理组大肠杆菌数量显著降低,低、中、高剂量处理组乳酸杆菌数量显著增加,低、高剂量处理组双歧杆菌数量显著增加,差异均有统计学意义(P〈0.05)。空白组血浆中TC[(5.18±0.87)mmol/L]、TG[(1.90±0.21)mmol/L]和LDL[(0.63±0.02)mmol/L]均高于对照组[分别为TC(2.54±0.39)mmol/L、TG(1.12±0.20)mmol/L、LDL(0.35±0.05)mmol/L],空白组HDL[(2.15±0.14)mmol/L]水平比对照组[(2.48±0.26)mmol/L]低,差异均具有统计学意义(P〈0.05)。与空白组相比,低、中、高剂量处理组的TC分别显著性降低,中、高剂量处理组的TG含量显著降低,低、中、高剂量处理组LDL显著降低,中、高剂量处理组HDL显Objective To explore the effect of polysaccharides from Sargassum graminifolium (SGP) on the intestinal flora of obese mice and the relationship between changes of the intestinal flora and biochemical indicators of blood lipid.Methods Sixty c57BL/6J mice were randomly divided into the conventional feed group (the control group, n=10) and the high-fat feed group(n=50). The model of obesity was established by using the animals in the high-fat feed group. With rejection of 10 animals that were not conformed to the selection standards, the remaining 40 mice were divided into the blank control group and the treatment group. The treatment group was subdivided into the low-dosage group that was given SGP at a dosage of 50 mg/kg by gavage, the medium-dosage group that was given SGP at a dosage of 100 mg/kg by gavage, and the high-dosage group that was given SGP at a dosage of 200 mg/kg by gavage, each consisting of 10 animals. After 8 weeks of feeding the SGP by gavage, detections were performed in biochemical indicators of blood lipid and the number of colonies of Escherichia coli, lactobacillus and bifidobacterium in the contents of the appendix.Results The number of colonies of lactobacillus [(7.05±0.15)lg CFU/g] and that of bifidobacterium [(8.75±0.41)lg CFU/g] in the blank control group was significantly decreased, as compared with that of the control group [(8.31±0.12)lg CFU/g and(10.17±0.31)lg CFU/g]. The number of colonies of Escherichia coli in the blank control group [(6.87±0.25)lg CFU/g] was significantly higher, as compared with that of the control group [(5.83±0.28)lg CFU/g], and statistical significance could be seen, when comparisons were made between them(P〈0.05). When compared with that of the blank control group, the number of colonies of Escherichia coli in the medium-dosage and high-dosage groups were significantly decreased.
