南方水稻黑条矮缩病毒P8蛋白的定位和致病性分析

摘　　要：P8是南方水稻黑条矮缩病毒(Southern rice black-streaked dwarf virus,SRBSDV)编码的一种外壳蛋白,该蛋白的其他功能未知。利用马铃薯X病毒(Potato virus X,PVX)异源病毒载体和pEarleyGate101在本氏烟(Nicotiana benthamiana)中表达了P8,通过荧光共聚焦显微镜观察这些蛋白的亚细胞定位情况,分析该蛋白的表达对本氏烟的影响。结果显示:P8主要定位于细胞质,在细胞质中形成颗粒状聚集体;与PVX空载体侵染的本氏烟相比,P8蛋白的表达能够促使本氏烟表现出一定程度的花叶和新叶皱缩症状,推测可能参与病毒的致病性过程。P8 was one of the major coat proteins of southern rice black-streaked dwarf virus(SRBSDV)and its other functions were poorly understood.In order to explore whether P8 involves the viral pathogenicity,P8 was exploited by expressing in Nicotiana benthamiana employing the vector Potato virus X(PVX)and p Early Gate101,respectively.The protein subcellular localization was observed by fluorescence confocal microscopy,and the effect of its expression on Nicotiana benthamiana is analyzed.The results showed that P8was localized in the cell cytoplasm and can form granular aggregates;P8 could induce leaf mosaic and new leaves shrivel symptoms when it was expressed by using PVX.So P8 might be involved in the pathogenic process of SRBSDV.
