摘  要:目的观察活血熄风法对偏头痛大鼠血浆P物质(SP)、神经激肽A(NKA)、内皮素(ET)含量的影响,探讨活血熄风法预防偏头痛的作用机制。方法将84只wistar大鼠随机分为空白组,模型组,镇脑宁组,西比灵组,活血熄风颗粒低、中、高剂量组,各组大鼠于末次给药1 h后开始制备偏头痛模型,除空白组外其他各组按照10 mg/kg皮下注射硝酸甘油注射液,空白组给予生理盐水10 mg/kg皮下注射。采用数码录像机定位拍摄的形式记录大鼠爬笼、挠头次数及采用酶联免疫法测定血浆中SP、NKA、ET的含量。结果在前三个时段(30 min、60 min、90 min),与空白组比较,模型组大鼠爬笼次数明显增多;在第四时间段(120 min),模型组与空白组比较爬笼次数无明显差异。在各时间段空白组偶有1次挠头动作,而在每个时间段模型组与空白组比较均具有明显的挠头动作;与模型组比较,西比灵组,活血熄风颗粒中、高剂量组挠头动作明显减少。与空白组比较,模型组大鼠血浆中SP、NKA含量升高,血浆中ET含量降低,差异有统计学意义(P<0.05);与模型组比较,大鼠服用镇脑宁、西比灵和活血熄风颗粒后均可使血浆中SP、NKA含量降低(P<0.05),ET含量升高(P<0.05),其中活血熄风颗粒低剂量组ET含量升高与模型组比较,差异无统计学意义(P>0.05)。活血熄风颗粒低、中、高剂量组与镇脑宁、西比灵组比较,以及活血熄风颗粒低、中、高剂量组3组间比较,差异均无统计学意义(P>0.05)。结论活血熄风颗粒可以通过调节血浆中SP、NKA、ET含量来改善大鼠的偏头痛症状,从而达到预防偏头痛的效果。Objective To observe the impacts on behavioristics and plasma SP, NKA and ET content in migraine rat models treated with the therapy for activating blood circulation and removing wind and explore the effect mechanism of this therapy. Methods Eighty - four wistar rats were randomized into a blank group, a model group, a zhennaojing group, a flunarizine group and a low - dose group, a middle - dose group and a high - dose group of huoxue xifeng granules. In I h after the final medication, migraine model was prepared in each group. Except the blank group, nitroglycerin injection was given subcutaneously, 10 mg/kg in each of the rest groups. Physical saline was used subcutaneously, 10 mg/kg in the blank group. The digital video recorder was used to film the frequency of rat climbing cage and scratching head. ELISA was used to determine the levels of SP, NKA and ET in plasma. Results In 30 min,60 min and 90 rain, compared with the blank group ,the frequency of climbing cage was increased apparently in the model group. In 120 min,the difference in the frequency of climbing cage was not apparent between the model group and the blank group. Scratching heat happened occasionally in each time point in the blank group. In comparison between the model group and the blank group,scratching head was more apparent in each time point. Compared with the model group, the frequency of scratching head was reduced apparently in the flunarizine group and the middle - dose group and the high - dose group of huoxue xifeng granules. Compared with the blank group, SP and NKA lev- els in plasma were increased in the model group and ET level was reduced, indicating the significant differences ( P 〈 0.05 ). Compared with the model group, the levels of SP and NKA were reduced after using zhenn- aoning,flunarizine and huoxue xifeng granules in the rats ( P 〈 0.05 ) and ET was increased ( P 〈 0.05 ), in which, the difference was not significant in ET increasing between the lowdose group and the model group ( P 〉 0.05 ).
