摘 要:[目的]确定铁皮石斛花中总黄酮超声辅助提取最佳工艺条件,并对其抗氧化活性进行评价。[方法]采用正交试验方法,以总黄酮提取率为考察指标,对铁皮石斛花总黄酮提取方法的影响因素进行探讨,并对铁皮石斛花总黄酮提取工艺进行优化。以羟自由基清除率、DPPH和ABTS自由基清除率为指标,对铁皮石斛花总黄酮的体外抗氧化活性进行评价。[结果]铁皮石斛花总黄酮的最佳提取条件:乙醇体积分数80%,料液比1:50,提取温度55℃,提取次数2次,在以上条件下,总黄酮提取率达1.99%。铁皮石斛花总黄酮对羟自由基、DPPH和ABTS自由基有较强的清除作用,且羟自由基抗氧化活性与总黄酮含量呈正相关。[结论]利用正交法分析结果可靠,得到了铁皮石斛花总黄酮超声提取的最佳工艺条件,且实验结果还表明铁皮石斛花具有较强的抗氧化活性。[Objective] To explore the optimal ultrasonic-assisted extraction conditions of total flavonoids from D-endrobium officinale flower and evaluate the antioxidant activity. [Methods] Orthogonal experiment was conducted to study the influence factor of extracting total flavones from Dendrobium offieinale flower,the yield of total flavones as inspection index,and then optimized the extraction technology. The antioxidant effects of the total flavones were determined by hydroxyl, DPPH and ABTS radical scavenging capacities. [Results] The optimal extraction eonditions of total flavonoids were ethanol concentration of 80% ,material-liquid ratio of hS0,extraetion temperature of 55℃ and 2 extraetion times. Under the optimal conditions,the maximum extraction rate of total flavonoids was 1.99%. Extracted total flavonoids had strong scavenging eapacity on hydroxyl free radicals, DPPH and ABTS free radicals. [Conclusion] Orthogonal experiment is applicable for the optimization of uhrasonie extraction of total flavonoids from Dendrobium offieinale flower. The total flavonoids from Dendmbium officinale flower have strong antioxidant capacity.
