摘 要:目的用亲水性的聚乙二醇对壳聚糖进行改性,探索分光光度法对聚乙二醇-壳聚糖聚合物中聚乙二醇接枝率的检测方法。方法通过化学方法合成亲水性mPEGCOONSu活化物,将其连接壳聚糖的氨基侧链上,得到改性了的聚乙二醇-壳聚糖接枝产物;用直接和间接两种分光光度法检测聚乙二醇-壳聚糖接枝产物的接枝率,并用核磁共振方法辅助证明。结果 ~1H-NMR确认了聚乙二醇-壳聚糖接枝的合成,测得接枝率为15.69±2.38%,直接间接两种方法测定接枝率分别为17.25±2.56%和16.34±1.98%。结论采用分光光度法测定聚乙二醇-壳聚糖的接枝率是一种简便、可行、有效的测定方法。Objective: Chitosan was modified with hydrophilic polyethylene glycol, and exploring the method of determination of the grafting ratio of chitosan-graft-poiyethylene glycol by Spectrophotometry. Method: Chitosan-grafi-polyethylene glycol was obtained through grafting hydrophilic mPEG spacer to the amino group of chitosan. The grafting ratio of chitosan-graft-polyethylene glycol was determined by direct and indirect spectrophotometry, and proved by nuclear magnetic resonance method. Results: Through t H-NMR experiment, the grafting ratio of chitosan-graft-polyethylene glycol was 15.69~2.38 %. The grafting ratio of chilosan-grafl-polyethylene glycol was 7.25~2.56 % and 16.34~1.98 % by direct and indirect spectrophotometry method. Conclusion: It is a simple, feasible and eft'cctive method to determine the grafting ratio of chitosan-graft-polyethylene glycol by spectrophotometry.
