逍遥丸对调节雄激素致排卵障碍模型大鼠卵巢氧化损伤和颗粒细胞凋亡的研究

摘　　要：目的观察逍遥丸对雄激素致排卵障碍模型大鼠卵巢氧化损伤和颗粒细胞凋亡的影响。方法 60只9日龄SD雌性大鼠随机选取20只为正常组,余40只皮下注射丙酸睾丸酮建立雄激素致排卵障碍模型大鼠模型,造模成功后,随机分为模型组和逍遥丸治疗组,每组20只,给药5周,于动情前期断头取血,比色法测定3组大鼠血清和卵巢T-AOC水平,硫代巴比妥法测定3组大鼠血清和卵巢MDA水平;免疫组化SP法检测3组大鼠卵巢组织中Caspase-3蛋白的表达。结果与正常组比较,模型组大鼠血清和卵巢中T-AOC水平显著降低,MDA水平显著升高(P<0.05),模型组卵巢颗粒细胞Caspase-3蛋白表达明显高于正常组(P<0.01)。与模型组比较,逍遥丸治疗组大鼠血清和卵巢中T-AOC水平升高,MDA水平降低(P<0.05),逍遥丸治疗组大鼠卵巢颗粒细胞Caspase-3蛋白表达值显著低于模型组(P<0.05)。结论逍遥丸能够改善排卵障碍疾病。Objective To observe the effects of Xiaoyao pill（ XYP） on ovarian oxidative damage and apoptosis of granulosa cells in androgen-induced sterile rats（ ASR）. Methods Forty female SD rats（ 9-day age） were randomly divided into control group（ n = 20） and model group（ n = 40）. The rat models with ovulation failure（ ASR） were established by subcutaneous injection with testosterone propionate,after modeling,the 40 rats were randomly divided into model control group and Xiaoyao pill treatment group（ medication for 5 weeks）,with 20 rats in each group. Then these rats were sacrificed and blood specimens were taken,the levels of T-AOC in serum and ovarian tissue were detected by colorimetry,and the levels of MDA in serum and ovarian tissue were detected by penthiobarbital method（ TBA）,and the expression levels of Caspase-3protein were detected by immunohistochemisty. Results As compared with those in control group,the levels of T-AOC in serum and ovarian tissue in model group were significantly decreased,however,the levels of MDA were significantly increased（ P 0. 05）. The expression levels of Caspase-3 protein in ovary granulosa cells were significantly higher than those in control group（ P 0. 01）. As compared with those in model control group,the levels of T-AOC in serum and ovarian tissue in Xiaoyao pill treatment group were significantly increased,however,the levels of MDA were significantly decreased（ P 0. 05）,moreover,the expression levels of Caspase-3 protein of ovary granulosa cells in Xiaoyao pill treatment group were significantly lower than those in model control group（ P 0. 05）. Conclusion XYP can improve follicular development and ovulation failure.
