摘  要:目的观察稳心颗粒对压力超负荷大鼠心肌肥厚和QT离散度以及缝隙连接蛋白43(Cx43)表达的影响。方法取45只雄性SD大鼠,以完全随机法分为正常组(N组)、心肌肥厚组(H组)、稳心颗粒组(W组)。采用腹主动脉缩窄法构建心肌肥厚大鼠模型,给予稳心颗粒6.25 g/(kg·d)治疗12周后,行超声心动图检测,并测定大鼠QT离散度,处死大鼠后以HE染色观察心肌病理形态变化,免疫组化法检测左室心肌Cx43蛋白的表达及分布,逆转录-链式聚合酶法检测心肌细胞Cx43 mRNA的表达。结果创建了压力超负荷诱导的心肌肥厚大鼠模型,超声心动图检测显示,与N组比较,H组大鼠出现明显心肌肥厚;W组和H组比较,以上参数有所改善。此外,H组的QT离散度比N组明显增加,而经稳心颗粒干预后的W组QT离散度较H组降低。同时,与N组大鼠比较,H组可见心肌细胞肥大、排列紊乱,且左室心肌组织Cx43的蛋白和mRNA表达明显减少;而W组和H组比较,心肌细胞排列紊乱则有所改善,Cx43的蛋白和mRNA的表达亦明显上调。结论稳心颗粒可改善压力超负荷大鼠的心肌肥厚,并降低QT离散度和改善心肌Cx43的重构。Objective TO observe the effects of Wenxin Granules on cardiac hypertrophy and QT dispersion and the expression of connexin43 (CX43) in pressure-overloaded rats. Methods Forty-five male Sprague-Dawley rats were divid- ed into normal group (group N), hypertrophy group (group H) and Wenxin Granules group (group W) by completely randomized method. The rat model of cardiac hypertrophy was established by constriction of abdominal aorta. When treated with Wenxin Granules 6.25 g/(kg·d) for 12 weeks, these rats were detected by echocardiography and QT dispersion. The pathological morphology of ventricle tissue was observed by HE staining, the expression of Cx43 protein and Cx43 mRNA in left ventricle was respectively detected by immunohistochemistry and RT-PCR after rats were sacri-ficed. Results The rat model of cardiac hypertrophy induced by pressure overload was established. Compared with group N, the rats in group H had obvious hypertrophy. Compared with group H, the above parameters were improved in group W. In addition, the QT dispersion Of group H was significantly higher than that of group N, and the QT dispersion of group W was lower than that of group H. Meanwhile, compared with group N, group H showed hypertrophy and disorder of cardiac muscle cell, and the expression of Cx43 protein and mRNA in left ventricle myocardium was significantly decreased; while the hypertrophy and disorder were improved in group W when compared with group H, Cx43 protein and mRNA expression was also significantly increased. Conclusion Wenxin Granules can improve cardiac hypertrophy in pressure-overloaded rats, decrease QT dispersion and improve the remodeling of Cx43 in myocardium.
