摘 要:目的:研究不同浓度川芎水提取物对缺氧人脐静脉内皮细胞(EAHY926)的作用。方法:体外培养EAHY926细胞,采用物理结合化学诱导缺氧建立模型,用川芎水提取液干预后,测定细胞活力,并检测乳酸脱氢酶、超氧化物歧化酶及一氧化氮(NO)的变化。结果:川芎嗪水提取液高、中剂量组均能显著提高细胞活力,降低乳酸脱氢酶外漏量,增加NO含量,升高超氧化物歧化酶活性(P<0.01)。结论:川芎水提取物能显著拮抗缺氧对EAHY926的损伤,其机制可能是清除自由基,增加NO含量,减少缺氧损伤。Objective:To study the protective effects of the water extract of Ligusticum chuanxiong on the hypoxic umbilical human vein endothelial cells (EAHY926). Methods:The EAHY926 cell was cultured in vitro, and the hypoxic model of which was establised using physical and chemical methods. After the hypoxic cells was interfered by the water extract of Ligusticum chuanxiong, the viability of cells was determined, and the levels of lactate dehydrogenase ( LDH), superoxide dismutase ( SOD ) and nitric oxide ( NO ) were detected. Results: High and middle dose water extract of Ligusticum chuanxiong could significantly improve the cell viability, reduce LDH leakage,and increase NO content and SOD activity (P 〈 0.01 ). Conclusions:The water extract of Ligusticum chuanxiong can significantly antagonize the hypoxia injury of EAHY926, the mechanism of which may involve in removing free radical, increasing NO content and reducing the hypoxic injury.
