摘  要:为明确酿酒葡萄浆果生长发育过程中白藜芦醇(Res)的变化及其相关酶活性以及底物苯丙氨酸的变化规律,探究葡萄生长发育过程中Res累积代谢的酶学机制,以酿酒葡萄赤霞珠(Vitis vinifera L.Cabernet Sauvignon)为试验材料,测定浆果发育过程中Res质量比的变化及Res合成相关酶:苯丙氨酸裂解酶(PAL)、肉桂酸-4-羟化酶(C4H)、4-香豆酸-辅酶A连接酶(4CL)、白藜芦醇合酶(RS)的活性以及底物L-苯丙氨酸质量浓度(L-Phe)的变化规律以及它们之间的相关性.在赤霞珠葡萄果实发育过程中,苯丙氨酸质量浓度呈现曲线增长,有2个关键转折点,在花后40和75 d左右;PAL、C4H、4CL、RS的活性均有2个高峰,在花后40和87d;Res呈现双峰值变化,峰值分别在花后40与87 d,成熟期其质量比迅速下降;苯丙氨酸质量浓度的变化与Res呈弱正相关,相关系数为0.690 7;PAL、C4H、4CL、RS活性变化与Res呈强正相关,相关系数分别为0.9366、0.893 9、0.832 0、0.983 8,均达到极显著水平;PAL、C4H、4CL、RS 4个酶活性之间呈极显著正相关,可能激活机制相同.Res质量比的变化与PAL、C4H、4CL、RS活性及底物L-Phe质量浓度的变化密切相关,存在伴随性,均具有2个高峰,分别在花后40与87 d左右,即Res合成受PAL、C4H、4CL、RS 4个酶协同调控以及底物LPhe的影响,其中RS为最关键的调节酶.To clarify variation of reseveratrol (Res) and related activity, variation of substrate phenylalauine during the different growth stage of wine grape berry. To probe the mechanism of res cumulative metabolic enzyme during growth stage of berry, to improve the theoretical for regulation the content of Res of the grape. Taking the fruit of Vitis vinifera L. Cabernet Sauvignon grape as experimental materials, measure the Res content variation and the activities of relevant enzymes during growth stage of berry, such as phenylalanineammonialyase (PAL), cinnamic acid -4- hydroxylase (C4H), 4-coumaric acid - coenzyme A ligase (4CL), activities of resveratrol synthase (RS) and the content of phenylalanine, further research variation and the relationship. The result shows that phenylalanine was a growing trend , has two peaks , 40 d and 75 d after flowers around; PAL ,C4H, 4CL , RS, Res showed double-peak variation , respectively 40 d and 87 d after flowers around, it is content rapid deline in maturity; the phenylalanine content showed an weakly significantly positive correlation with the Res, the correlation coefficient of 0.6907; There existed a good positive correlation between the activity of PAL, C4H, 4CL, RS and Res, and the correlation coefficient respectively was 0.9366, 0.8939, 0.8320 and 0.9838, reaching an extremely remarkable level, including RS was highest; the activity of PAL,C4H,4CL,RS were significantly positive correlation with the same activation mechanism. The content variation of Res in Vitis vinifera L. Carbernet Sauvignon are closely related to the activity and substrate of PAL, C4H, 4CL, RS with concomitant and two peaks , respectively 40d and 87d after flowers around, that Res synthesis by PAL, C4H, 4CL, RS coordinately regulated and L-Phe influenced, RS was the most important one.
