摘  要:胆固醇是维持细胞膜脂筏稳定结构重要组分。一些囊膜病毒感染过程依赖胆固醇。研究利用Real Time PCR和病毒滴度试验,分析细胞膜和病毒囊膜胆固醇是否影响鸡传染性支气管炎病毒(IBV)感染鸡胚肾(CEK)细胞。结果表明,胆固醇萃取剂甲基-β-环糊精去除细胞膜胆固醇后,可抑制IBV感染CEK细胞并呈剂量依赖性,补充外源性胆固醇后,IBV感染力部分恢复,说明IBV感染依赖细胞膜胆固醇;去除细胞膜胆固醇可影响IBV感染侵入和释放过程。去除病毒囊膜胆固醇对IBV感染力无显著影响。IBV感染CEK细胞时依赖细胞膜胆固醇,而非病毒囊膜胆固醇。Cholesterol is an important component to maintain the stable structure of lipid rafts on cellular membrane. For some enveloped viruses, the infection of host cell depends on the participation of cholesterol. In this paper, pretreatment of chicken embryo kidney cells (CEKs) with methyl-^-cyclodextrin (M/3CD), a drug used to deplete cholesterol from membranous structure, to analyze whether the cellular or viral membrane cholesterol played a role in chicken infectious bronchitis virus (IBV) infection of CEKs. The results showed that significantly decrease of IBV infection in a dose-dependent manner through real time RT-PCR and virus titer test, and IBV infection could be partially restored by supplementing exogenous cholesterol, indicated that IBV infection depended on cellular membrane cholesterol. Further studies discovered that the removal of cellular membrane cholesterol affected IBV attachment and release stage.However, the removal of viral envelope cholesterol had no effect on IBV infection. These results demonstrated that IBV infection of CEKs was dependent on cellular membrane and no viral envelope cholesterol.
