摘  要:枯草芽孢杆菌是生产微生物菌肥的有效微生物菌种之一,芽孢含量直接影响菌肥效果。为降低生产成本,提高发酵液中芽孢数量,探究牛粪沼液对枯草芽孢杆菌(Bacillus subtilis)D1产孢影响。采用分批培养法确定发酵培养基中沼液最适浓度及碳源与氮源种类和含量。利用比浊法和稀释平板涂布法检测枯草芽孢杆菌的菌体数量及芽孢产率,采用火焰原子吸收光谱法检测沼液中金属离子含量。结果表明,牛粪沼液中含有对产孢有显著影响的Ca^(2+)、Mn^(2+)、Mg^(2+)三种离子;当沼液与水配比为11时,产孢效果最佳;麸皮和大豆粉分别是菌株生孢最佳碳源、氮源;最适碳源和氮源含量分别为2.0%和0.5%(质量比),35℃,120 r·min^(-1)恒温培养24 h后,产芽孢率可达83.77%。Bacillus subtilis is one of the effective microorganism strains producing bacterial manure, and the spore content is one of the important factors directly affect the effect of bacterial manure.In order to improve the spore number in the fermented liquid, and reduce the production cost, the influence of biogas slurry of cow dung on spore production of Bacillus subtilis D1 was investigated. The optimal concentration of biogas slurry and the types and contents of the carbon source and nitrogen source in the fermentation medium were determined using batch cultivation method. The number of bacteria and the rate of spore production were detected with turbidimetric method and the dilution plate coating methodl respectively. And the contents of metal ions in the biogas slurry were determined by the flame atomic absorption spectrometry. The results showed that the biogas slurry of cow dung contained three kinds of ions Ca2+, Mn2+, and Mg2+, which had the significant effects on the sporulation. The rate of spore production were the best when the ratio of biogas slurry to the water was 1:1. The optimal carbon and nitrogen sources were bran and soybean meal, respectively. The optimal concentration of carbon source and nitrogen source were 2.0% and 0.5, The highest rate of spore production was 83.77% in optimal medium at 120 r.min-1 and 35℃ after 24 h incubation.
