摘 要:目的:探讨咳喘方对支气管哮喘模型大鼠气道重塑的干预作用。方法:将30只SD雄性大鼠分为对照组、模型组、中药组,每组10只。除对照组外均行支气管哮喘造模;中药组予咳喘方灌胃,其余两组予0.9%Na Cl注射液灌胃,疗程3周。采用免疫组化法检测各组大鼠肺组织转化生长因子-β1(TGF-β1)的表达;并取肺组织经HE染色进行病理观察,图像分析测定其气道壁总面积(Wat)、管壁平滑肌面积(Wam)、支气管底膜周径(Pbm)、支气管周围胶原纤维组织面积(Wac)的数值。结果:模型组TGF-β1表达较对照组明显升高(P<0.01);中药组TGF-β1计数较模型组明显降低(P<0.01)。模型组Wat、Wam、Wac水平较对照组明显升高(P<0.01);中药组Wat、Wam、Wac水平较模型组明显降低(P<0.01)。结论:咳喘方可抑制支气管哮喘大鼠肺组织TGF-β1表达和气管壁增厚。Objective:To discuss the effects of Kechuan Decoction on airway remodeling in asthmatic rats.Methods:Thirty male SD rats were divided into the control group,model group and Chinese medicine group,10 rats in each group.Except the control group,bronchial asthma model was established in other rats.The Chinese medicine group was orally treated with Kechuan Decoction,the other two groups were arally treated with 0.9% Nacl,with a course of 3 weeks.The expression of transforming growth factor-β_1(TGF-β_1) in lung tissue of rats in each group was detected by immunohistochemistry,and the lung tissue was collected for pathological observation after HE staining,the numerical value of airway wall area(Wat),smooth muscle area(Wam),bronchial basement membrane perimeter(Pbm) and collagenous tissue area(Wac) were detected by image analysis.Results:The TGF-β_1 level in the model group was obviously increased compared with the control group(P〈0.01);the TGF-β_1 level in the Chinese medicine group was obviously decreased compared with the model group(P〈0.01).The levels of Wat,Wam and Wac in the model group were obviously increased compared with the control group(P〈0.01);the levels of Wat,Wam and Wac in the Chinese medicine group were obviously decreased compared with the model group(P〈0.01).Conclusion:Kechuan Decoction can inhibit TGF-β1expression and tracheal wall thickening in asthmatic rats.
