摘  要:目的:观察升降散对糖尿病大鼠双歧杆菌、大肠杆菌及IL-6的影响,探讨其作用机制。方法:取70只SD雄性大鼠,随机取10只作为正常对照组,余60只用高脂饲料喂养配合小剂量链脲佐菌素诱导建立糖尿病模型,取成模大鼠50只分为模型对照组、拜糖平组、升降散低剂量组、升降散中剂量组和升降散高剂量组,共分6组,每组10只,各组按相应药物剂量干预6周后,每组取8只大鼠样本采用Elisa法检测IL-6、INS,Realtime PCR技术进行细菌定量测定。结果:治疗前,模型对照组、拜糖平组、升降散低、中、高三个剂量组较正常对照组空腹血糖(FBG)、IL-6、INS及大肠杆菌数量均明显升高,但双歧杆菌数量明显减少;治疗前后,正常对照组及模型对照组组内各指标无明显变化。治疗后,拜糖平组及升降散低、中、高剂量FBG在2周、4周及6周均逐渐降低,且高于同时间正常对照组,低于同时间模型对照组(P<0.05);IL-6、INS、大肠杆菌数量较模型对照组明显减少,但同时高于正常对照组(P<0.05);双歧杆菌数量较模型对照组、正常对照组明显增多(P<0.01)。尤以升降散中剂量组增加双歧杆菌数量、降低大肠杆菌数量,抑制炎症因子、降低胰岛素含量效果明显。结论:升降散能通过增加益生菌数量,降低致病菌数量达到调整肠道菌群结构的目的,从而抑制炎症状态,降低血糖。其部分作用机制可能是通过调整肠道菌群、抑制炎症因子而减轻胰岛素抵抗(IR),增加胰岛素利用率实现的。Objective To observe the effect and the mechanism of Chinese herbal medicine on Bifidobacterium, Escherichia coli and IL -6 in diabetic rats. Methods 70 SD male rats, 10 rats as normal control group, only more than 60 high fat diet combined with low dose of streptozotocin induced diabetic model was established, the model 50 rats were divided into model group, Bai Tangping group, Shengjiangsan low dose group, middle dose group Shengjiangsan Shengjiangsan and high dose groups were divided into 6 groups, 10 rats in each group, each group according to the corresponding dose of drug for 6 weeks, 8 rats from each group of samples was detected by Elisa IL-6, INS Realtime, PCR technology for quantitative determination of bacteria. Results Before treatment, model group and acarbose group, Shengjiangsan low, medium and high dose groups compared with normal control group, fasting blood glucose (FBG), IL -6, INS and the number of Escherichia coli were significantly increased, but the number of bifidobacteria was significantly reduced; before and after the treatment, the normal control group and model control group each index in the group had no obvious change. After treatment, acarbose Shengjiangsan group and low, middle and high dose of FBG in 2 weeks, 4 weeks and 6 weeks were gradually decreased, and higher than that of the normal control group, compared with model control group ( P 〈 0. 05 ) ; IL - 6, INS, the number of Escherichi- a eoli than the model control group was significantly reduced, but at the same time higher than the normal control group ( P 〈 0. 05 ) ; the number of bifidobacteria compared with model control group, normal control group increased significantly ( P 〈 0. 01 ) . Especially in the middle dose group, the number of bifidobacteria increased, the number of E. coli decreased, the inflammatory factors and insulin content were decreased significantly.
