银杏叶滴丸对冠心病患者TLR4/NF-κB信号通路及免疫指标的影响

摘　　要：目的评价银杏叶滴丸对冠心病患者TLR4及NF-κB信号通路及免疫相关指标的影响。方法 100例患者随机分为试验组50例和对照组50例。两组患者均给予常规西药治疗,试验组同时给予银杏叶滴丸,1次5丸,1日3次,连续30天。采用酶联免疫吸附实验(ELISA)测定两组患者血浆超敏-C反应蛋白(hs-CRP)、白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)含量;采用免疫比浊法测定免疫球蛋白IgA、IgG、IgM水平;采用实时荧光-PCR仪检测Toll样受体4(TLR4)、单核细胞核因子-κB(NF-κB)的表达。结果治疗后,两组TLR4、NF-κB的表达量较治疗前显著降低(P<0.05),观察组显著低于对照组(P<0.05);观察组hs-CRP、IL-6、TNF-α的含量治疗后较治疗前显著降低(P<0.05),且治疗后均显著低于对照组(P<0.05);治疗后,两组IgA、IgE、IgM的水平较治疗前显著降低(P<0.05),试验组显著低于对照组(P<0.05)。结论银杏叶滴丸治疗冠心病的作用机制可能与抑制外周血TLR4/NF-k B炎症信号通路的激活,抑制炎症反应并改善机体免疫功能紊乱有关。OBJECTIVE To investigate the effect of Ginkgo Leaf Dripping Pill on TLR4/NF-κB inflammatory signal pathway and immune function of patients with coronary heart diseases. METHODS One hundred patients with coronary artery diseaseswere randomly divided into observation group（ n = 50） and control group（ n = 50）. The patients in two groups were treated with some medicine,which on the basis of symptomatic treatment. However,the patients in observation group,on the basis of the control group,were treated with Ginkgo Leaf Dripping Pill,with a treatment course of 30 days for both groups. The expression levels of TLR4 and NF-κB mRNA in peripheral blood were detected by Real time-PCR. The serum levels of IL-6,hs-CRP,TNF-αwere measured by ELISA. The immunoglobulins（ IgA,IgG,IgM） were examined by turbidimetric inhibition immuno assay for both groups. RESULTS After 30 days treatment,compared with those before treatment,the expression levels of TLR4 and NF-κB,IL-6,hs-CRP,TNF-α,IgA,IgG,IgM were significantly decreased after treatment for observation group（ P〈0. 05）,and the expression levels of TLR4 and NF-κB,IgA,IgG,IgM were significantly decreased after treatment for control group（ P〈0. 05）. What＇s more,compared with control group,the expression levels of TLR4 and NF-κB,IL-6,hs-CRP,TNF-α,IgA,IgG,IgM of observation group were significantly decreased（ P〈0. 05）. CONCLUSION Ginkgo Leaf Dripping Pill can significantly inhibit the activation of TLR4/NF-κB inflammatory signal transduction pathway and can improve immunological function disorders,which may be one of the action mechanisms of atorvastatin in treatment of coronary heart diseases.
