摘  要:策勒黑羊具有全年发情、多胎的特点,是新疆策勒县特有的优良绵羊品种。为研究与保护策勒黑羊遗传资源,为后续开展逆转录酶聚合酶链式反应(RT-PCR)、斑点杂交试验和构建c DNA文库试验等提供条件,试验采用经典异硫氰酸胍(Trizol)法提取策勒黑羊卵巢总RNA,通过核酸蛋白仪测定所提取RNA在不同波长下的吸光度值,计算RNA的提取量与纯度,并利用琼脂糖凝胶电泳显示提取结果和验证RNA的完整性。结果表明:采用Trizol法提取得到的策勒黑羊卵巢RNA的浓度、纯度与完整性均可达到后续试验的要求,可为后续策勒黑羊种质资源保护和利用等生物学试验的开展提供参考依据。The Qira black sheep has the characteristics of annual estrus and multiple births, and it is a specific and excellent breed of sheep in Qira County of Xinjiang. To study and protect the genetic resources of the Qira black sheep and provide the conditions for the subsequent development of reverse transeriptase polymerase chain reaction (RT- PCR), dotblot hybridization test and cDNA library testing, a classical guanidinlum isothiocyanate method was used to extract the total RNA from the ovary of Qira black sheep with Trizol Reagent. The light absorption values of the extracted RNAs Under different Wavelengths were determined by :protein nucleic acid analyzer, The extraction amount and purity of RNAs were calculated, and the agarose gel electrephoresis was used to show the extraction results and validation of RNA integrity. The results showed that the concentration, purity and completeness of the ovarian RNA of Qira black sheep that extracted with TrizoI Reagent could meet the requirements of subsequent experiments, which could provide reference basis for subsequent development of biological experiments such as the protection and utilization of germplasm resources of Qira black sheep.
