摘  要:目的探讨5-羟色胺2C(5-HT2C)受体对新生大鼠离体延髓脑片基本节律性呼吸放电(RRDA)的调节作用。方法取2日龄Sprague-Dawley大鼠18只制备离体延髓脑片标本,使用人工脑脊液(ACSF)灌流脑片,吸附电极记录脑片舌下神经根RRDA。观察使用空白ACSF灌流脑片15、30、45、60 min时RRDA的吸气时程(TI)、呼吸周期(RC)和放电积分幅度(IA)变化情况;使用含1、5、10、20μmol·L^(-1)5-HT2C受体激动剂MK212的ACSF灌流脑片后RRDA的TI、RC和IA变化情况;使用含1、5、10、20μmol·L^(-1)5-HT2C受体阻断剂RS102221的ACSF灌流脑片后RRDA的TI、RC和IA变化情况。结果空白ACSF灌流脑片15、30、45、60 min时RRDA的TI、RC和IA比较差异均无统计学意义(P>0.05)。1μmol·L^(-1)MK212的ACSF灌流脑片后RRDA的TI、IA和RC与给药前比较差异均无统计学意义(P>0.05);5μmol·L^(-1)MK212的ACSF灌流脑片后RRDA表现为TI延长、IA增加、RC缩短(P<0.05);10μmol·L^(-1)MK212的ACSF灌流脑片后RRDA表现较5μmol·L^(-1)MK212的ACSF灌流脑片后TI进一步延长、IA进一步增加、RC进一步缩短(P<0.05);但20μmol·L^(-1)MK212的ACSF灌流脑片后与10μmol·L^(-1)MK212的ACSF灌流脑片后比较,RRDA的TI、IA、RC差异均无统计学意义(P>0.05)。1μmol·L^(-1)RS102221的ACSF灌流脑片后RRDA的TI、IA和RC与给药前比较差异均无统计学意义(P>0.05);5μmol·L^(-1)RS102221的ACSF灌流脑片后RRDA表现为TI缩短、RC延长(P<0.05),IA差异无统计学意义(P>0.05);10μmol·L^(-1)RS102221的ACSF灌流脑片后RRDA表现较5μmol·L^(-1)RS102221的ACSF灌流脑片后TI缩短、IA减少、RC延长(P<0.05);但20μmol·L^(-1)RS102221的ACSF灌流脑片后与10μmol·L^(-1)RS102221的ACSF灌流脑片后比较,RRDA的TI、IA、RC差异均无统计学意义(P>0.05)。结论 5-HT2C受体对延髓呼吸中枢起着正性调节作用。Objective To explore the regulatory effect of 5-hydroxytryptamine 2C( 5-HT2C) receptor on the rhythmic respiratory discharge activity( RRDA) of medulla slice of neonatal rat in vitro. Methods Eighteen Sprague-Dawley rats( 2days old) were taken to prepare the medullary slices specimen. The medullary slices were perfused with artificial cerebrospinal fluid( ACSF),and the RRDA of sublingual nerve root was recorded by suction electrode. The inspiratory time( TI),respiratory cycle( RC) and integral amplitude( IA) of RRDA of medulla slices at the time points of 15,30,45,60 min after being perfused blank ACSF were observed. The TI,RC and IA of RRDA of medulla slices after being perfused ACSF which contained 1,5,10,20 μmol·L^-15-HT2 C receptor agonist MK212 respectively were observed. The TI,RC and IA of RRDA of medulla slices after being perfused ACSF which contained 1,5,10,20 μmol · L^-15-HT2 C receptor agonist RS102221 respectively was observed. Results There was no statistic difference in the TI,RC and IA of RRDA of medulla slices among the time points of15,30,45,60 min after being perfused blank ACSF( P〉0. 05). There was no statistic difference in the TI,RC and IA of RRDA of medulla slices before and after being perfused 1 μmol·L^-1MK212 ACSF( P〉0. 05); the TI,RC and IA of RRDA of medulla slices after being perfused 5,10,20 μmol·L^-1MK212 ACSF were significantly bigger than before and after being perfused 1 μmol· L^-1MK212 ACSF( P〉0. 05); the TI,RC and IA of RRDA of medulla slices after being perfused 10,20 μmol·L^-1MK212 ACSF were significantly bigger than after being perfused 5 μmol·L^-1MK212 ACSF( P〈0. 05); there were no statistic difference in the TI,RC and IA of RRDA of medulla slices after being perfused 10 μmol·L^-1MK212 ACSF and 20 μmol·L^-1MK212 ACSF( P〈0. 05). There was no statistic difference in the TI,RC and IA of RRDA of medulla slices before and after being perfused 1 μmol·L^-1RS102221 ACSF( P〈0. 05); the TI,RC of RRDA
