摘  要:利用生物信息学方法筛选辐照处理后肺腺癌A549细胞中的差异表达lncRNAs,从Gene Expression Omnibus(GEO)公共数据库获得辐照后肺腺癌A549细胞的基因表达芯片GSE24876,利用GEO2R软件分析差异表达基因,利用DAVID在线数据库对差异基因进行重注释获取差异表达的lncRNAs。结果共筛选出相关的差异表达lncRNAs 33个,与未辐照处理的A549细胞相比,有4个表达上调,29个表达下调,差异均具有统计学意义。利用Gene-E软件进一步分析和验证lncRNAs的表达情况,结果与GEO2R的分析基本一致,但需进一步实验验证。预后分析的结果表明,高表达LINC00319(HR 1.21[1.02~1.42],p=0.026)和LINC01095(HR1.18[1.01~1.40],p=0.042)的肺癌患者拥有较差的总生存期。To identify the key differentially expressed long non-coding RNAs in irradiated lung carcinoma A549 cells by bioinformatics analysis, the mRNA microarray dataset GSE24876 was downloaded from the Gene Expression Omnibus database and analyzed by GEO2R to obtain differentially expressed gene probes. Re-annotation of probes was performed for the differentially expressed genes using DAVID. In total, 33 differentially expressed lncRNAs were identified from GSE24876, which included 4 upregulated lncRNAs and 29 downregulated lncRNAs in irradiated A549 cells compared with non-irradiated A549 cells. The Gene-E software was then used to confirm the expression levels of the lncRNAs and the results were similar to those analyzed by GEO2R. Among them, high expression of LINC00319(HR 1.21 [1.02~1.42],p= 0.026) andLINC01095(HR 1.18 [1.01~1.40], p=0.042) was associated with worse overall survival in lung cancer patients. However, further studies are needed to verify the results of this study.
