半夏厚朴汤对抑郁症型大鼠的治疗作用研究

摘　　要：目的建立抑郁症大鼠模型,并研究半夏厚朴汤对大鼠抑郁症的治疗作用及作用机制,为临床防治抑郁症提供实验和理论依据。方法将SD大鼠12只随机分为四组:正常组(3只),模型组(3只),半夏厚朴汤组(3只),西药氟西汀组(3只)。后三组进行电击足底、通宵照明、禁水或禁食、换笼饲养、昼夜颠倒等,各种刺激随机选用,制造抑郁症大鼠模型。分别用半夏厚朴汤、氟西汀和生理盐水给对应抑郁症大鼠模型灌胃。用水迷宫、敞箱实验、糖水试验等分别测试正常组、模型组与药物组大鼠的反应。最后断头取血,取海马,测试海马中乙酰胆碱变化,并根据西医发病机制筛选特异性较高的指标,进行多重比较研究。对比分析得出半夏厚朴汤对抑郁症大鼠的影响。结果半夏厚朴汤组一般状况(皮色、精神、饮食等)优于对照组大鼠;半夏厚朴汤组大鼠在糖水试验中变现的认知状况优于对照组;半夏厚朴汤组海马重量大于其他组大鼠。且血清乙酰胆碱和海马乙酰胆碱含量分析高于模型组和氟西汀组。结论半夏厚朴汤对抑郁症大鼠具有治疗作用,且效果优于西药氟西汀。Objective To establish the depression rat model, and to study the therapeutic effeet and the mechanism of Banxia Houpu decoction on rats of depression, to provide experimental and theoretical basis for clinical prevention and treatment of depression. Methods 12 SD rats were randomly divided into four groupsi normal group （3 rats）, model group （3 rats）, banxia Houpu decoction group （3 rats） and fluoxetine group （3 rats）, The three groups were electric shock foot, night lighting, water deprivation or fasting, for cage rearing, day and night, and so on, a variety of stimuli randomly selected, manufacturing depression rat model. Respectively with fluoxetine and saline banxiahoupu decoction, corresponding model rats with depression. The water maze, open field test and sucrose consumption test were tested in the normal group, model group and drug group rat reaction. Finally, the blood was taken, the hip- pocampus was taken to test the changes of acetylcholine in the hippocampus, and the specificity of the higher index was selected ac- cording to the pathogenesis of Western medicine. Comparative analysis of the effect of Banxia Houpu decoction on depression in rats was carried out. Results The general condition of Banxia Houpu decoction group （skin color, spirit, diet, etc.） was better than that of the control group. Banxia Houpu decoction group rats were realized in sucrose test in cognitive status was better than the control group. Banxia Houpu decoction group was greater than the weight of other groups of rats＇ hippocampus, The levels of acetylcholine and acetylcholine in serum were higher than that in the model group and the fluoxetine group, Conclusion Banxia Houpu decoction has a therapeutic effect on depression in rats, and the effect is better than Western medicine fluoxetine.
