摘  要:目的:研究丹酚酸B(SAB)对转化生长因子-β1(TGF-β1)诱导大鼠粘连组织成纤维细胞(ATF)增殖活性和胞外基质(ECM)合成的影响。方法:采用Cell Count Kit法(CCK-8)检测SAB对TGF-β1诱导ATF增殖影响。实时荧光定量PCR技术(Real-time PCR)、酶联免疫吸附法(ELISA)检测SAB对TGF-β1诱导ATF I型胶原蛋白(COLI)、纤维连接蛋白(FN)、金属基质蛋白酶-9(MMP-9)和金属基质蛋白酶抑制剂-1(TIMP-1)mRNA和蛋白表达的影响。结果:10 ng·m L^(-1)TGF-β1可显著提高细胞外基质成分COLI和FN的表达。10μmol·L^(-1)SAB可以抑制TGF-β1诱导ATF增殖活性(P<0.05),并显著下调COLI和FN的表达;另外还可促进MMP-9表达、降低TIMP-1活性,调节细胞外基质降解平衡。结论:SAB预防术后腹膜粘连的机制与抑制TGF-β1诱导的ATF增殖,调节MMP-9和TIMP-1分泌平衡促进ECM降解有关。Objective: To study the effects of salvianolic-acid B( SAB) on proliferation and extracellular matrix( ECM) synthesis in rat adhesive tissue fibroblasts( ATF) induced by transforming growth factor beta-1( TGF-β1). Methods: The proliferation of ATF was detected by Cell Count Kit( CCK-8). The gene expressions of collagen I( COLI),fibronectin( FN),matrix metalloproteinase-9( MMP-9) and tissue inhibitor of metalloproteinase-1( TIMP-1) were detected by real-time PCR. ELISA was employed to determine the protein levels of COLI,FN,MMP-9 and TIMP-1. Results: TGF-β1 of 10 ng·m L-1could effectively up-regulate the COLI and FN expressions.SAB of 10 μmol·L-1could suppress the ATF proliferation( P 〈 0. 05) and down-regulate the expressions of COLI and FN in TGF-β1-stimulated ATF. Besides,SAB could also significantly up-regulate MMP-9 while down-regulate TIMP-1 in TGF-β1-stimulated ATF. Conclusion: SAB can inhibit TGF-β1-induced ATF proliferation. It can also restore the balance of MMP-9 and TIMP-1 expression in ATF,thus improve the ECM degradation,which may be the preventive and prophylactic mechanisms of SAB on post-operative adhesion.
