摘  要:目的引进并运用逆转录多聚酶链式反应-限制性片段长度多态性分析方法(RT-PCR-RFLP方法)快速鉴别2015年宜春市麻疹野病毒感染病例与疫苗株病毒引起的相关麻疹病例。方法运用realtime RT-PCR对宜春市400例疑似麻疹病例咽拭子标本进行麻疹病毒核酸检测,引进RT-PCR-RFLP方法对麻疹病毒核酸检测阳性咽拭子标本进行检测是否含有AflⅡ酶切位点(疫苗株病毒有,野病毒没有)鉴别疫苗相关麻疹病例。结果麻疹病毒核酸检测阳性标本为9例,阳性率为2.25%;9例麻疹病毒核酸检测阳性标本RT-PCR-RFLP方法检测的结果为均未能被AflⅡ酶切开,为麻疹野病毒感染。结论成功引进和建立宜春市麻疹野病毒感染病例与疫苗株病毒引起的相关麻疹病例的RT-PCR-RFLP快速鉴别方法,并鉴别了2015年宜春市9例麻疹病例均由麻疹病毒野病毒感染引起。Objective To establish and apply reverse transcription polymerase chain reaction-restriction fragment length poly- morphism analysis (RT-PCR-RFLP method) for rapidly identifying wild measles virus strains and vaccine strains in Yiehun City in 2015. Methods The RT-PCR assay was used to detect cases of suspected measles cases throat swab specimens for the virus nu- cleic acid detection. RT-PCR-RFLP was adopted to identify wild strains and vaccine strains by determining the existence of en- zyme-containing All I1 restriction site (vaccine strains,not wild strain). Results Nine cases of positive measles virus specimens were detected,accouting for 2.25%. RT-PCR-RFLP identification result was as follows :The strain showed a single band,which means it can not be digested by Aft 1I. Conclusion A method for rapid identification of Yichun City of wild measles virus strains and vaccine strains was introduced,and identified nine cases of Yichun City in 2015 suspected measles cases caused by wild measles virus infection.
