江苏地区2株山羊痘病毒的分离与鉴定

摘　　要：为对江苏部分地区羊场羊痘病毒进行有效的监测和防控,及时了解病毒遗传进化趋势,降低规模化养羊业的风险,本研究从江苏部分地区养羊场采集疑似羊痘病羊的皮肤丘疹、水疱或脓疱组织病料样品,分别接种9日龄SPF鸡胚绒毛尿囊膜、胎羊睾丸细胞(LT)和牛肾细胞(MDBK),观察鸡胚病变和细胞病变情况。同时通过PCR方法扩增羊痘病毒p32基因进行序列分析,并进行p32基因限制性片段长度多态性分析。结果显示,接种鸡胚绒毛尿囊膜形成典型的痘斑,胚体出血;系统进化分析显示,两株分离株属于山羊痘病毒(GTPV);限制性片段长度多态性分析结果也显示符合GTPV的特征。本研究所分离到的2株GTPV,为当地制定山羊痘的防控策略提供依据。In order to effective monitoring and control pox virus in parts of Jiangsu, the lesion tissue samples were collected from the affected goat in the farms, and the suspensions of clinical samples were prepared from skin papules, blisters, or pustule tissues of sick goats, and inoculated into 9-day-old chick embryo membranes, LT cells and MDBK cells for virus isolation, respectively. The P32 gene of goat pox virus was amplified by PCR, and sequenced for analysis. The results showed that the typical acne marks and embryo hemorrhage were produced in the chick embryo chorioallantoic membranes, and the cytopathic effects were induced in the LT cells and MDBK cells after the inoculation. Phylogenetic analysis showed that virus isolates belonged to the goat pox virus. Furthermore, restriction fragment length polymorphism analysis also confirmed that the isolates possessed the characteristics of goat pox virus. The isolation goat pox virus woul facilitate for the development of local prevention and control strategies of goat pox virus.
