摘  要:目的通过对比三种不同方法致大鼠心脏骤停心肺复苏(CPR)后脑组织中细胞间黏附因子-1(ICAM-1)和血管细胞黏附分子-1(VCAM-1)的表达,从而判断复苏后的脑损伤情况。方法利用窒息、3妇重物压迫胸部、夹闭心底血管丛三种方法制造雄性清洁级SD大鼠心脏骤停动物模型,分别在复苏成功后3、6、12h处死,取大脑皮层组织用免疫组化方法测定ICAM-1、VCAM-1的蛋白表达,并采用SPSS17.0统计软件进行统计学分析。结果与对照组比较,实验组各组各相应时间点的ICAM-1和VCAM-1的表达均明显增高(P〈0.05),但各实验组之间ICAM-1和VCAM-1的表达差异无统计学意义(P〉0.05)。3kg重物组6h和12h脑组织ICAM-1和VCAM-1含量较3h高(P〈0.05),夹血管组6h和12hICAM-1表达较3h高(P〈0.05)。结论三种不同方法所致的大鼠脑损伤的程度基本一致。三种模型均可用于今后的CPR后脑损伤的研究。Objective To determine the brain injury degree of rats by comparing the expres- sions of ICAM - 1 and VCAM - 1 in brain tissue after cardiopulmonary resuscitation from cardiac arrest induced by three different methods. Methods Cardiac arrest animal models of rats were built with the use of three methods including suffocation, chest oppression with 3 - kilogram object, and clamping of cardiac plexus. Three groups of rats were respectively sacrificed 3, 6 and 12 hours after resuscitation. The cerebral cortical tissues were harvested to determine ICAM - 1 and VCAM - 1 expression by immu- nohistochemical methods, Results Compared with the control group, ICAM - 1 and VCAM - 1 ex- pressions at the corresponding time points in the experimental groups were significantly increased (P 〈 0.05), but the differences in ICAM - 1 and VCAM - 1 expression among the experimental groups were not significant (P 〉 0.05). ICAM - 1 and VCAM - 1 contents in brain tissue of rats sacrificed 6 h and 12 h after resuscitation from cardiac arrest induced by chest oppression with 3 - kilogram were higher than in rats sacrificed 3 h after resuscitation (P 〈 0.05); for rats resuscitated from cardiac arrest induced by the clamping of cardiac plexus, ICAM - 1 expressions in brain tissue of rats sacrificed 6 h and 12 h after resuscitation were higher than in rats sacrificed 3 h after resuscitation (P 〈 0.05). Conclusion The degree of brain injury caused by three different methods are the basically same. All the three models built by different cardiac arrest - inducing methods can be used for the research on brain injury after cardiopul- monary resuscitation.
