摘 要:目的研究黄芪提取物(EA)对老年性痴呆(AD)大鼠核转录因子-κB(NF-κB)和NF-κB抑制蛋白(IκB-a)蛋白的影响。方法大鼠海马内注射Aβ25-35制备AD大鼠模型,实验分4组:对照组、模型组、黄芪组和西药治疗组。黄芪组和西药治疗组分别用黄芪提取物和脑复康灌胃,对照组和模型组用等量生理盐水灌胃。用Morris水迷宫测定各组大鼠的学习记忆能力,Western blot测定各组大鼠海马NF-κB、IκB-a和caspase-3蛋白表达水平。结果模型组的逃避潜伏期延长,单位时间内跨越原平台次数减少,NF-κB和caspase-3蛋白表达升高,而IκB-a蛋白表达降低,与对照组比较均有统计学差异(P<0.01);黄芪组的逃避潜伏期缩短,单位时间内跨越原平台次数增多,NF-κB和caspase-3蛋白表达降低,而IκB-a蛋白表达升高,与模型组比较均有统计学意义(P<0.05或P<0.01),其作用效果与西药治疗组相当(P>0.05)。结论黄芪提取物通过提高AD大鼠海马区IκB-a的含量,使游离NF-κB的含量减少,激活caspase-3受到抑制,从而抑制了神经细胞的凋亡。Objective To study the effects of astragalus extract(EA)on the protein expression of nuclear factor-κB(NF-κB)and IκB-a in rats with Alzheimer's disease(AD).Methods The AD model rats were established by injecting Aβ25-35 into districts of hippocampuses.The rats were divided into 4 groups:control group,model group,EA group,piracetam group.The rats in EA group and piracetam group were treated with EA and piracetam lavage respectively,with isodose physiological saline lavage in control group and model group.The ability of learning and memory was detected by Morris water maze.The protein expression level of NF-κB,IκB-a and caspase-3 in the hippocampus was detected by western blot.Results Compared with control group,the incubation period of avoiding was lengthened,the frequency of passing through platform was decreased,the expression level of NF-κB and caspase-3 was increased,while the expression level of IκB-a was decreased in model group(P <0.05).Compared with the model group,the incubation period of avoiding was obviously shorten,the frequency of passing through platform was increase,the expression level of NF-κB and caspase-3 was decreased,while the expression level of IκB-a was increased in EA group(P <0.05 or P <0.01).Its effect was similar with western medicine treatment group(P >0.05).Conclusion EA can increase the content of IκB-a and decrease the content of free NF-κB in the hippocampus of AD rat,and restrained the activation of caspase-3,inhibit the apoptosis of nerve cells.
