摘  要:【目的】研究左旋咪唑(LMS)刺激的猪内皮细胞(VEC)对单核源树突状细胞(MoDC)的抗原递呈功能的影响。【方法】通过LMS刺激猪血管内皮细胞后,筛选出能够使内皮源IL-8表达量上调的低质量浓度LMS(10^(-6) mg/mL),将经LMS处理的内皮细胞与MoDC共培养,收集共培养的MoDC。流式细胞术检测MoDC的CD80/86和MHC-II的表达;丝裂霉素C处理后的MoDC与淋巴细胞混合,MTS法和流式细胞术分析MoDC抗原递呈和对淋巴细胞转化的影响。共培养分为诱导后共培养和诱导共培养。【结果】LMS处理VEC后,两种共培养方式所得MoDC的MHC-II与CD80/86阳性率、淋巴细胞刺激指数、Tc和Treg细胞阳性率均显著下调,而Th细胞阳性率和CD4^+/CD8^+比值显著上调。【结论】低浓度LMS处理的VEC可抑制MoDC的抗原递呈能力。[Objective]The aim of this paper is to study the effect of porcine endothelial cells(VEC) stimulated with levamisole (LMS) on antigen presenting function of monocyte-derived dendritic cells (MoDC). [Methods] The low concentration of LMS (10-6mg/mL) which can up-regulated expression of endothelial IL-8 was selected. MoDCs were collected after LMS-treated VEC co-cultured with MoDC and the expressions of CD80/86 and MHC-II in MoDC were assayed by flow cytometry. The mit- omycin C-treated MoDC were mixed with lymphocytes and MTS assay and flow cytometry were employed for the analysis of antigen presentation and effect of MoDC on lymphocyte transformation. Two methods of co-culture included post-induced co- culture and induced co-culture. [Results]The expression of MHC-II and CD80/86 of MoDC, stimulation index of T cell, and the positive rates of Tc and Treg were significantly down-regulated in LMS-induced VEC co-cultured by the two methods of af- ter. The positive rates of Th and ratio of CD4+/CD8+ were up-regulated significantly.[Conclusion] VEC stimulated with low concentration of LMS could inhibit the antigen presenting ability of MoDC.
