摘 要:目的:在前期文献研究与临床研究基础上,观察玉竹水煎液对力竭运动大鼠心肌的保护作用,研究其作用机制。方法:SD大鼠40只,体重180~220g,雌雄各半,随机分为5组,分别是:(1)安静对照组;(2)力竭运动组;(3)低剂量力竭组;(4)中剂量力竭组;(5)高剂量力竭组。实验期间除安静组外,采用为期4周的游泳训练,1次/天,60分/次,5天/周;3~5组大鼠,每天灌胃以相应高、中、低浓度的玉竹水煎液,1~2组给予等量的生理盐水;实验第28天,2~5组大鼠一次性游泳至力竭,(1)分别记录各组游泳至力竭时间;(2)联合检测血清中CK-MB、cTn1、AST心肌酶谱含量;(3)检测血清MDA、GSH-px和SOD等抗氧化指标;(4)电镜观察心肌超微结构的变化。结果:(1)玉竹能延长大鼠运动至力竭的时间,并呈现一定的剂量依赖关系;(2)力竭运动提高了血清中cTnⅠ、CK-MB、AST的含量,但玉竹补充可以降低其含量,且与剂量成正相关,高剂量组效果最佳;(3)力竭运动降低了血清中GSH-px和SOD的含量,升高了MDA的含量,玉竹可以改善这一效果,并呈剂量依赖性;(4)电镜观察发现:力竭运动引起心肌超微结构改变,表现为大部分心肌细胞肌丝溶解、线粒体肿胀、肌膜损伤、部分细胞器外溢等变化,玉竹高中低剂量组对这一现象都有一定的保护作用。结论:(1)力竭运动导致血清中cTnⅠ、CK-MB、AST的升高,玉竹可以缓解力竭运动对心肌酶谱的影响,并且剂量越高,效果越好;(2)力竭运动显著降低血清中GSH-px和SOD的含量,升高MDA的含量,玉竹可以增强机体抗氧化酶的含量,减少自由基的产生,从而降低氧化应激引起的心肌损伤,呈剂量依赖性。Based on the former literature reviews and clinic research,explore the protective function and mechanism of Polygonatum odoratum,(Mill.)Druce on the myocardium of the mice that experienced exhaustive exercise.Method:20 female and 20 male mice(weight range:180~220g)were randomly selected into 5 group:(1)control group;(2)exhaustive exercise group;(3)low-dose medicine combined exhaustive exercise group;(4)middle-dose medicine combined exhaustive exercise group;(5)high-dose medicine combined exhaustive exercise group.Except the control group,the mice in other four groups participated into 4-week swimming training,60 minutes per time,once per day,5 days per week.In three medicine groups(3,4,5 group),drugs were chronically administered to animals,according to the different levels of doses,while the mice in 1 and 2 groups were provided normal saline.On Day 28,the mice in 2,3,4,5 groups experienced exhaustive-exercise,recording exercise time,detecting the level of serum cTnI,CK-MB and AST myocardial enzyme content,detecting the level of serum MDA,GSH-px and SOD antioxidant,and observing ultra-structural changes of myocardium via electron microscopy.Findings:(1)Polygonatum odoratum might extend the time for getting exhaustive,showing dose-response relationship.(2)Exhaustive exercise can increase the level of serum CK-MB,cTn1 and AST myocardial enzyme content.However,Polygonatum odoratum appeared the positive association in the capabilities for preventing these variable increase.By the way the highest dose group exhibited the best outcomes.(3)Exhaustive exercise reduced serum GSH-px and SOD content,increasing the content of MDA,while,based on the dose,Polygonatum odoratum can act as the positive role,accompanying with dose-dependence.(4)Electron microscopy showed:exhaustive exercise induced changes of myocardial ultra-structure,as most myo-filament myocardial cells dissolution,mitochondrial swelling,muscle membrane damage,some organelles spillover and other changes,
