摘 要:目的探讨经大鼠腹部皮下植入医用几丁糖对其免疫系统的影响。方法将96只Wistar大鼠随机分为医用几丁糖高剂量组(5 ml)、低剂量组(1 ml)、假手术组(5 ml无菌生理盐水)、空白对照组(未处理)、免疫抑制组(取材前连续4 d经大鼠腹腔注射环磷酰胺)和免疫刺激组(取材前12 h单次腹腔注射脂多糖),根据考察周期将每组16只大鼠再随机分为1、4、8和12周4个实验操作小组。对大鼠以下指标进行检测:免疫器官系数、免疫器官组织病理学观察、NK细胞杀伤活性、T淋巴细胞亚群分析和T/B淋巴细胞增殖情况。结果医用几丁糖植入后1、4、8和12周,医用几丁糖高剂量组及低剂量组大鼠的免疫器官系数、NK细胞杀伤活性、T淋巴细胞亚群分析、T/B淋巴细胞增殖的结果与假手术组和空白对照组比较,差异均无统计学意义(P>0.05),免疫器官组织病理学观察未见异常;免疫抑制组大鼠的免疫器官系数、NK细胞杀伤活性、T淋巴细胞亚群分析、T/B淋巴细胞增殖的结果与其他各组比较,差异均有统计学意义(P<0.05),免疫器官组织病理学观察可见淋巴细胞数目大量减少等免疫抑制现象;免疫刺激组大鼠的脾脏系数、T淋巴细胞亚群分析、T淋巴细胞增殖的结果与其他各组比较,差异均有统计学意义(P<0.05),免疫器官组织病理学观察可见淋巴细胞数目大量增多等免疫刺激现象。结论经大鼠腹部皮下植入医用几丁糖后,未对其免疫系统产生明显影响。Objective To investigate the effect of implantation of medical chitosan into abdominal subcutaneous tissue on immune system of rats. Methods Ninety-six Wistar rats were randomly divided into six groups. The rats in high dose medical chitosan, low dose medical chitosan and sham-operation groups were treated with 5 ml medical chitosan, 1 ml medical chitosan and 5 ml physiological saline respectively, while those in immunosuppression group were injected i. p. with cyclophosphamide 4 d before samples were taken, those in immunostimulation group were injected i. p. with lipopolysacchardie (LPS) 12 h before samples were taken, and those in blank control group were untreated. The sixteen rats in each group were further divided into four subgroups, four for each, then weighed and killed 1, 4, 8 and 12 weeks after treatment, of which the immune organ coefficient, histopathology, killing activity of NK cells, T-lymphcocyte subset and proliferation of T / B lymphocytes were investigated. Results The immune organ coefficient, killing activity of NK cells, T lymphcocyte subset and proliferation of T/B lymphocytes of rats in high dose medical chitosan group 1, 4, 8 and 12 weeks after implantation showed no significant difference with those in sham operation and blank control groups (P 〉 0. 05), while no abnormality was observed in histopathology. However, immune organ coefficient, killing activity of NK cells, T lymphcocyte subset and proliferation of T / B lymphocytes of rats in immunosuppression group showed significant difference with those in other groups (P 〈 0. 05 ), while immunosuppression such as remarkable decrease of lymphocytes were observed in histopathological examination. The spleen coefficient, T lymphocyte subset and T lymphocyte proliferation of rats in immunostimulation group showed significant difference with those in other groups (P 〈 0. 05), while immunostimulation such as remarkable increase of lymphocytes were observed in histopathological examination.
