摘 要:目的通过RT-PCR及荧光原位杂交方法观察胰岛素样生长因子-1(IGF-1)在正常股骨头与缺血坏死性股骨头组织中的表达差异,探讨IGF-1在股骨头坏死中的发病机制。方法取股骨粗隆间骨折及缺血坏死性股骨头行人工股骨头置换或全髋人工关节的正常股骨头、缺血坏死性股骨头骨组织各10例;分别取组织2~5g放入冷冻管存入液氮用于荧光原位杂交及IGF-1基因的RT-PCR。结果 IGF-1的荧光原位杂交图像显示正常骨组织中的表达量比坏死组织增多,RTPCR的2-ΔΔCt平均值分别为0.58±0.34;0.43±0.19;正常股骨头骨组织中IGF-1基因RT-PCR的2-ΔΔCt平均值与缺血坏死性股骨头骨组织平均值相比,差异有统计学意义(P<0.05)。结论IGF-1基因在正常与缺血坏死性股骨头骨组织表达不同,IGF基因表达量的减少可能是股骨头缺血坏死的重要原因之一。Objective To observe the different expressions of insulin-like growth factor 1 (IGF 1) in the bone tissues of normal and ischemic and necrotic femoral heads by RT-PCR and fluorescence in situ hybridization method and to explore the pathogenesis of IGF - 1 in femoral head necrosis. Methods 10 cases of normal bone tissues and 10 cases of ischemic and necrotic bone tissues were removed from the femoral heads of intertrochanteric fractures and artificial femoral head replacement or total hip joint replacement. 2-5 g of bone tissues were placed in liquid nitrogen freezing tube to be used for fluorescent in situ hybridization and IGF gene RT-PCR. Results The fluorescence in situ hybridization images of IGF-1 showed that the expressions of normal bone tissues were more than those of the necrotic tissues and that the average values of IGF-1 gene RT-PCR2^-△△Ct were 0.58±0.34 and 0.43±0.19 respectively( P 〈0.05). Conclusion IGF-1 gene has different expressions in the bone tissues of normal and ischemic and necrotic femoral heads and the decreased IGF gene expressions may be one of the important causes of ischemic necrosis of the femoral head .
