摘　　要：目的通过荧光原位杂交及RT-PCR实验方法观察骨形态发生蛋白-2(BMP-2)在正常与缺血坏死性股骨头骨组织中的表达差异,探讨BMP-2在两种骨组织中的变化规律。方法取股骨粗隆间骨折及缺血坏死性股骨头需行人工股骨头置换或全髋人工关节置换的正常股骨头、缺血坏死性股骨头骨组织患者各8例;分别取组织2~5 g放入冷冻管存入液氮用于荧光原位杂交及BMP-2基因的RT-PCR。结果BMP-2的荧光原位杂交图像显示正常骨组织中的表达量比坏死组织增多,RT-PCR的2-ΔΔCt平均值分别为0.55±0.35,0.45±0.20;正常股骨头骨组织中BMP-2基因RT-PCR的2-ΔΔCt平均值与缺血坏死性股骨头骨组织平均值相比P<0.05。结论 BMP-2基因在正常与缺血坏死性股骨头骨组织有统计学意义,因此BMP-2基因表达量的减少也许是股骨头缺血坏死的重要原因之一。Objective By fluorescence in situ hybridization and rt-pcr method to observe the BMP-2（BMP-2） expression differences in the normal and ischemic and necrotic femoral head bone, BMP-2 variation in two kinds of bone tissue. Methods Intertrochanteric fractures and ischemic and necrotic femoral head requires artificial femoral head replacement or total hip joint replacement of normal femoral head, all the 8 patients with ischemic and necrotic femoral head bone tissue; respectively in group 2-5 g in liquid nitrogen freezing tube used in fluorescent in situ hybridization and BMP-2 gene RT-PCR. Results BMP-2 of fluorescence in situ hybridization images show that the amount of expression in normal tissue than the necrotic tissue increased, RT-PCR2-ΔΔCt average is（0.55±0.35） and（0.45±0.20）; in normal femoral bone BMP-2 gene RT-PCR2-ΔΔCt average compared with average ischemic and necrotic femoral head bone tissue P〈0.05. Conclusion BMP-2 gene in normal and ischemic and necrotic femoral head bone tissue was statistically significant, therefore decreases may BMP-2 gene expression is one of the important reasons of the ischemic necrosis of the femoral head.
