壮药排钱草总生物碱对大鼠肝星状细胞胶原和细胞因子生成的影响

摘　　要：目的:研究排钱草总生物碱对大鼠肝星状细胞(HSC-T6)相关胶原和细胞因子的分泌影响。方法:体外培养HSC-T6细胞,将排钱草总生物碱(终浓度为500、250、125、62.5、31.25、15.625μg/m L)作用于细胞48h后,以MTT法检测细胞存活率,求出半数抑制浓度(IC50),根据IC50选取无细胞度浓度进一步研究;收集细胞上清液,用ELISA法分别检测Ⅰ型胶原(COL-Ⅰ)、Ⅲ型胶原(COL-Ⅲ)、Ⅳ型胶原(COL-Ⅳ)、转化生长因子(TGF-β1)、血小板衍生因子(PDGF)的含量。结果:排钱草总生物碱能明显抑制HSC-T6细胞的增殖,其抑制作用与药物终浓度呈量效关系,求得IC50为337μg/m L;并显著降低HSC-T6细胞生成的COL-Ⅰ、COL-Ⅲ、TGF-β1,PDGF含量,与空白对照组比较,P<0.05或P<0.01。结论:排钱草总生物碱对HSC-T6细胞增殖有显著抑制作用,并能降低COL-Ⅰ、COL-Ⅲ、TGF-β1、PDGF的含量。Objective ：To study the effect of total alkaloids of Desmodium pulchellum on the HSC -T6 prolifera- tion and the content of collage andcytokine Method ： Culture HSC - T6 cells in vitro, taking the total alkaloids of Desmodium pulchellum （final concentration were 500,250,125,62.5,31.25,15. 625 μg/mL）act on the cell of HSC - T6 cell, adopting MTT colorimetric assay for cellsurvival rate after 48 hours and count IC50 ; The culture su- pernatant fluid was collected, the contents of COL - Ⅰ ,COL - Ⅲ ,COL - Ⅳ ,TGF - β1 ,PDGF were measured by ELISA. Result ： The total alkaloids of Desmodium pulchellum could inhibit HSC - T6 cell proliferation, and with the increase in drug concentration,the effect of inhibiting the more significant. The IC50 was 337ug/mL; can sig- nificantly decrease the level of the contents of COL - Ⅰ , COL - Ⅲ, TGF - β1, PDGF, P 〈 0.05 或 P 〈 0.01. Conclusion： The total alkaloids of Desmodium pulchellum inhibited the proliferation of HSC -T6 cell and de- crease the level of the contents of COL -Ⅰ , COL - Ⅲ ,TGF - β1 ,PDGF.
