摘  要:目的探究朗格汉斯细胞是否通过CD207参与皮肤对表皮葡萄球菌的免疫应答。方法利用朗格汉斯细胞缺失的CD207(Langerin)-DTA小鼠及野生型小鼠,分为对照组,胶带粘贴组及胶带粘贴后涂抹表皮葡萄球菌(staphylococcus epidermidis,SE)组。取小鼠皮损制备苏木精-伊红(HE)切片,分析表皮厚度及炎症;制备皮损单细胞悬液,流式细胞仪分析浸润细胞类型及朗格汉斯细胞特异性C型凝集素受体CD207对SE的结合。结果胶带粘贴后,小鼠表皮轻度增厚,但在CD207-DTA小鼠与野生型小鼠之间没有差异。在胶带粘贴后涂抹SE组,CD207-DTA小鼠表皮增厚程度及皮损浸润CD4阳性T细胞低于野生型小鼠。朗格汉斯细胞特异性C型凝集素受体CD207可结合表皮葡萄球菌。结论朗格汉斯细胞可能通过CD207参与皮肤对表皮葡萄球菌的免疫应答。Objective To explore whether Langerhans cells participate in the skin immune response to staphylococcus epidermidis through CD207.Methods CD207(Langerin)-DTA and wild-type mice were divided into the control, tape stripped, and tape stripped with application of staphylococcus epidermidis group.The skin lesions were harvested for hematoxylin-eosin(HE)staining to analyze epidermal thickness and inflammation.Single cell suspension of skin was used to assess the types of inflammatory cells, using flow cytometry.The binding of Langerhans cell specific C-type lectin receptor(CD207)to staphylococcus epidermidis was also determined by flow cytometry.Results Tape stripping caused mild increase in epidermal thickness in both CD207-DTA and wide-type mice.The application of staphylococcus epidermidis to the tape stripped skin induced a less extent of epidermal hyperplasia and CD4+ T cell infiltration in CD207-DTA vs.wild-type mice.In addition, CD207 could bind to staphylococcus epidermidis in vitro.Conclusion Langerhans cells may participate in the skin immune response to staphylococcus epidermidis via CD207.
