摘  要:采用果蝇三龄幼虫为试验材料,经过剥离唾腺、解离、漂洗、染色、压片处理后,通过显微镜观察染色结果。染色步骤中,使用卡宝品红染色法、孚尔根染色法及孔雀绿染色法三种染色方法对照醋酸洋红染色法设定四组试验。试验结果表明卡宝品红染色法、孚尔根染色法及孔雀绿染色法的染色效果比醋酸洋红染色法的染色效果均有所提升。综合考虑染色时间、实验操作、染色液的配置及染色效果四个方面,孔雀绿染色法染色时间少、实验操作简便、染色液配置简单、染色效果好,使用孔雀绿染色法效率最高,可用于教学实验中。Adopting the third instar larvae of Drosophila melanogaster as test material, this test obtainedthe dyeing results by observing microscopic after stripping salivary glands, dissociation, rinsing, dye-ing and tableting process. In the process of dyeing, four groups of test were designed by comparingthe carbol fuchsin dyeing, Feulgen dyeing and malachite green dyeing method with the carmine stai-ning dyeing. The results show that the carbol fuchsin dyeing, Feulgen dyeing and malachite greendyeing have been more effective than that of the carmine staining dyeing. Taken into the overall con-sideration of dyeing time, experimental operation, preparation of the dyeing and dyeing effects, themalachite green dyeing has the best efficiency with less time, simpler experimental operation andpreparation of dyeing and better dyeing effects, which can be applied in teaching experiments.
