姜黄素对人气道平滑肌细胞免疫调控功能影响的研究

摘　　要：目的研究姜黄素对体外培养的人气道平滑肌细胞(airway smooth muscle cells,ASMCs)分泌Th1、Th2细胞因子功能影响,为姜黄素对局部气道免疫调控提供理论依据。方法原代培养ASMCs,第4~6代细胞用于实验,细胞用不同浓度姜黄素(μmol/L)(4.24、8.48、16.97、33.93、67.86、135.73、271.45)诱导培养48 h,经细胞鉴定后,用ELISA法测定上清液中IL-4、IFN-γ浓度。结果姜黄素对ASMCs分泌IL-4、IFN-γ分泌水平有显著影响,与对照组对比,随浓度增高,加姜黄素8.48μmol/L以上浓度组,IL-4水平显著下降(pg/m L)(对照组68.05±13.37,实验组56.77±15.54(P<0.05),49.24±11.34,51.41±9.56,50.05±10.13,45.41±7.53,43.89±8.77(P均<0.01));67.86μmol/L以上浓度组,IFN-γ显著增高(pg/m L)(对照组:22.21±4.28,实验组:33.98±7.32,38.53±7.64,45.31±7.59,P均<0.01)。结论姜黄素能调控气道平滑肌细胞分泌IFN-γ、IL-4,调节Th1、Th2的平衡并向Th1方向偏移,可能在抑制局部气道过敏反应中有一定作用。Objective To study the effect of curcumin on the secretion of Th1 and Th2 cytokines in vitro cultured human airway smooth muscle cells （ASMCs）, and to provide a theoretical basis for the regulation of local airway immune regulation. Methods We separate ASMCs for primary culture and 4 6 generation cells were used in the experiment. The cells were cultivated with different concentrations of curcumin （μmol/L）（4.24, 8.48, 16.97, 33.93, 67.86, 33.93, 67.86） for 48 h. We measured the concentration of in supernatant fluid with ELISA method. Results Curcumin can affect the secretion of IL - 4, IFN-γ levels in ASMC s. Compared with the control group, with the concentration of curcumin increasing （over 8.48 μmol/L）, IL - 4 levels decreased significantly （pg/mL） （P〈0.01） （P〈0.01）, when concentration of curcumin is over 67.86 μmol/L ,IFN-γ, increased significantly （P〈0.01）. Conclusion Curcumin can regulate airway smooth muscle cells secreting IFN-γ, IL-4, Adjusting the balance of Th1/ Th2 and shifting to the Thl direction. There may be some role in inhibiting local airway allergic reactions.
