摘 要:[目的]观察揉髌手法对兔膝关节骨关节炎模型血清代谢组学的影响,探讨其治疗膝关节骨关节炎的可能作用机制。[方法]52只新西兰兔分为手法组、对照组、假模组和正常组各13只,手法组、对照组手术造成骨内高压型动物模型,手法组每只兔予揉髌手法治疗17次。造模后12周末耳缘静脉取血,核磁共振波谱仪检测血清样本,进行PLS-DA分析等。[结果]骨关节炎软骨组织病理学Mankin分级评分显示对照组中度退变,手法组、假模组轻度退变,正常组基本正常;PLS-DA得分图显示手法组与对照组术后12周末有分离趋势;载荷图提示手法组、对照组、假模组和正常组的差异性代谢物有Glucose、Lactate、Glycine等。[结论]揉髌手法延缓兔膝关节软骨组织的退变可能与葡萄糖、乳酸盐、甘氨酸等代谢物的作用有关。[Objective]To observe the effect of rubbing patella technique on serum metabolomics in osteoarthritis model at knee joints of rabbit and discuss the possible mechanism for treatment. [ Methods ] Fifty-two New Zealand rabbits were randomly divided into four gourps: manipulation group, model group, dummy-model group and the normal group, with 13 rabbits in each group. A high intraosseous pressure model was established in the manipulation group and model group. Rubbing patella technique was performed 17 times for each rabbit in the manipulation group. Twelve weeks after modeling, blood samples were taken from the ear vein and the serum samples were detected by nuclear magnetic resonance spectroscopy, and PLS-DA analysis was performed. [Results] The Mankin grading score of osteoarthritis cartilage histopathology showed moderate degeneration in the model group, mild degeneration in manipulation group and dummy-model group. There appeared no obvious change in normal group. Twelve weeks after operation, serum analysis showed that the data from the manipulation group and model group had a tendency to separate on PSL-DA scoring table. The loading diagram showed that the differences of metabolites for the manipulation group, the model group, the sum-model group and the normal group were glucose, lactate, glycine and so on.[ Conclusion ] Thedelay of degeneration inknee articular cartilage- of rabbits with rubbing patella technique may be related to the effects of metabolites such as glucose, lactate and glycine.
