免疫抑制剂干预对食蟹猴感染HBV的血清学影响

摘  要:[目的]研究免疫抑制剂干预对食蟹猴感染乙型肝炎病毒(HBV)的血清学影响。[方法]筛选健康的成年食蟹猴24只,随机分为空白对照组、氢化可的松组、环孢素组、环磷酰胺组,每组6只。空白对照组注射HBs Ag阴性正常血清,氢化可的松组、环孢素组和环磷酰胺组使用相应免疫抑制剂处理2次后开始接种HBV携带者血清,每日观察动物行为变化,干预4周后,取血样检测乙肝病毒全套、HBV-DNA、肝功能,并取肝脏组织行病理检测。[结果]空白对照组未见HBs Ag、HBs Ab、HBV-DNA阳性发生。环孢素组、环磷酰胺组中,部分动物检测出HBs Ag阳性,部分食蟹猴出现HBV-DNA阳性表达并持续,与空白对照组比较有统计学意义(P<0.05);感染动物均出现ALT、AST显著升高。[结论]经环孢素及环磷酰胺免疫抑制剂干预后可以提高食蟹猴感染HBV的成功率。[ Objective ] To study the serological effects of immunosuppressive agents on HBV Infection in cynomolgus monkeys. [Methods]Twenty-four healthy adult cynomolgus monkeys were randomly divided into control group, hydrocortisone group, cyclosporine group and cyclophosphamide group (Six monkeys for each group). The control group was injected with HBsAg negative normal serum. Hydrocortisone , cyclosporine and cyclophosphamide groups were treated with the corresponding immune intervention agent. After 2 times of treatment, HBV carriers were inoculated to observe the behavior change. After 4 weeks of intervention, blood samples were taken to detect the complete hepatitis B virus, HBV-DNA and liver function, and the liver tissues were conducted with pathological examination. [ Resuhs]HBsAg, HBsAb and HBV-DNA positives were not detected in the control group. HBsAg positive were detected in some monkeys in cyclosporine and cyclophosphamide groups. Some cynomolgus monkeys showed positive expression of HBV-DNA , which continued. It was statistically significant compared with that of control group (P〈0.05). ALT and AST were elevated in infected animals. [Conclusion]Immunosuppressive therapy can significantly improve the success rate of HBV infection in cynomolgus monkeys.
