摘 要:以纤维蛋白粗酶液为原料,用硫酸铵沉淀分离、离子交换层析、凝胶过滤层析、凝胶电泳分析等方法分离纯化纤维蛋白溶解酶Ⅰ,通过SDS-PAGE电泳检测纯度,通过抗凝血、血凝块溶解、溶血、纤维蛋白溶解酶酶源的激活及水解实验探讨其生物活性。结果表明,纯化后的纤维蛋白溶解酶Ⅰ呈现单一条带,该酶具有体外溶栓、抗凝血、溶解血凝块、激活纤溶酶原和水解纤维蛋白原的作用。This this study, the fibrinolytic enzyme Ⅰ is isolated and purified from the crude protein enzyme of the crude fiber and the ammonium sulfate precipitation, ion exchange chromatography, gel filtration chromatography and gel electrophoresis, purity is detected by SDS-PAGE eleetrophoresis, and the biological activity is studied by means of anticoagulation, blood clot lysis, hemolysis, activation of fibrinolytic enzyme and hydrolysis. Experiment results show that the purified fibrinolytic enzyme -Ⅰ show a single band, and the enzyme has the effects of thrombolysis, anticoagulation, dissolution of blood clots, activation of plasminogen and Hydrolyzing fibrinogen.
