摘 要:目的探讨两种绒毛细胞培养方法对绒毛细胞培养结果的影响以及其染色体核型分析在胚胎停育中的应用。方法取103例胚胎停育的绒毛细胞分别采用胰酶与胶原酶混合消化法和组织块直接培养法进行细胞培养、染色体核型分析。结果 103例绒毛组织两种方法均培养成功98例,培养成功率为95.1%,胰酶与胶原酶混合消化法比组织块培养法培养时间更短。共分析绒毛标本98例,发现异常核型42例,异常率42.86%,其中数目异常最多(39例)。结论胰酶与胶原酶混合消化法可以缩短绒毛细胞培养时间,绒毛染色体核型异常是胚胎停育的重要因素。Objective:To study the effect of two methods to the result of villus cultivate and discuss the application value of villus cells karyotype in the embryo damage. Methods:Villus cells from 103 women of the embryo damage were cultured for two methods:tissue block culture and cells cultured after Trypsin and collagenase digestion and prepared for karyotype analysis. Result:103 cases were cultured and 98 cases for two methods were all cultured successfully with the successful rate of 95.1%. The time of cell culture after rypsin and collagenase digestion was shorter than that of tissue block culture. Analysis of villus cell of 98 cases,42 cases were detected with adnormal karyotype,accounting for 42.86%. And adnormal number was the major type of adnormal karyotype(39 cases). Conclusion:The time of cell culture after Trypsin and collagenase digestion was shorter than that of tissue block culture.Embryo villus cell abnormal karyotype is one of the important factors of the embryonic damage.
