摘 要:为了了解乙烯信号受体在文心兰切花衰老中的作用,利用RACE技术克隆了文心兰乙烯受体基因OnERS1(Gen Bank登录号为KP410729)。生物信息学分析表明,OnERS1基因编码631个氨基酸,分子量为70 773.7,等电点为6.82,是跨膜蛋白,与其它植物的ETR乙烯受体蛋白的相似性在74%~99%之间,属ETR1类乙烯受体。基因表达分析结果表明,OnERS1基因在根和蕊柱中表达量最高,在叶片、侧瓣和花萼中次之,在茎和唇瓣中表达量最低;文心兰切花衰老期间OnERS1基因的表达量在第0~6 d持续增加,第6 d后突然下降,第8 d后下降到一个极低的水平。与此同时,花器官中的乙烯释放从第8 d之前缓慢上升,第8~10 d急剧升高。OnERS1基因的时空表达特异性表达说明它可能与花衰老有关。OnERS1(Gen Bank accession number is KP410729),a member of ethylene receptor gene family was cloned by RACE techniqued from Oncidium Gower Ramsey in order to understand the role of ethylene receptor gene in the senescence of cut flower of Oncidium.Bioinformatics analysis showed that OnERS1 gene a trans-membrane protein encoded 631 amino acids with a molecular weight of 70 773.7,an isoelectric point(IP)of 6.82.OnERS1 shared a similarity of 74%-99% among the ethylene receptor proteins of other plant species and belonged to the ETR1 subfamily.Gene expression analysis demonstrated that OnERS1 gene had the highest expression levels in roots and gynandria,followed by those in sepals,lateral petals and leaves,lowest in stems and lip petals;during the cut flower senescence,the expression levels in various flower organs continually increased to a maximum at 6 d,and then declined sharply.At the same time,ethylene production increased slowly until 8 d,and then dramatically increased.Temporal and spatial expression of the OnERS1 gene suggested that its function may be related with flower senescence.
